Effects Of Yanghe Pingchuan Granules On Airway Remodeling And Wnt/beta-catenin Signal Pathway In Chronic Asthma Model Rats

Objective:To study the effects of Yanghe Pingchuan granules on airway remodeling and Wnt/?-catenin signal pathway in rats with chronic asthma.Methods:Fifty male and clean Sprague-Dawley rats were randomly divided into normal group,model group,Yanghe Pingchuan group,inhibitor group and dexamethasone group,with 10 rats in each group.The rat model of chronic asthma was established by multi-point injection of ovalbumin combined with aluminum hydroxide and aerosol challenge of ovalbumin,and drug intervention was given for 6 weeks from the 20 th day of the experiment.Yanghe Pingchuan group was given intragastric administration of Yanghe Pingchuan granules according to the dose of 7.74 g/kg once a day,dexamethasone group was given intragastric administration of dexamethasone according to the dose of 0.125 mg/kg once a day,and the inhibitor group was given XAV939 1.75 mg/kg intraperitoneal injection every other day.The spirit,activity,coat color and death of rats in each group were observed and recorded.The pathological changes of lung tissue were observed by HE staining.The deposition of collagen fibers in airway epithelium of rats was observed by Masson trichrome staining.The levels of serum total Ig E,OVA specific Ig E,PDGF and TGF-?1 were detected by ELISA.The expression of ?-catenin,Axin and Cyclin D1 protein in lung tissue was detected by Westernblot,and the expression levels of Wnt-3a,Wnt-5a,?-catenin,Axin and Cyclin D1 m RNA in lung tissue were detected by RT-q PCR.Results:1.The general condition of rats in each group: the rats in the normal group were slightly restless during the atomization of normal saline,and their activities were normal,their spirit and activity were good,and the coat was smooth and shiny after atomization.The rats in each group had different degrees of restlessness during the aerosol challenge of ovalbumin,and all the rats lay still and did not move after the atomization of ovalbumin.In the model group,the excitation response was more obvious,the respiratory sound thickened obviously in the later stage,the phlegm sound could be heard gradually,the mental state was general,the coat color was basically the same as that of the normal group,and the body weight did not decrease significantly.The spirit and activity of the rats in the Yanghe Pingchuan group and inhibitor group were good,the hair color was slightly yellower than that in the normal group,and the body weight was slightly lower than that in the model group.In the later stage of intervention,the spirit of the rats in the dexamethasone group significantly deteriorated,the activity decreased,the hair color was yellow,and the body weight was significantly lower than that in other groups.In the late stage of intervention,3 rats in the dexamethasone group died,but there was no death in the other groups.2.Compared with the normal group,obvious inflammatory cell infiltration was observed in the airway and surrounding tissues of the model group,and the total thickness of airway wall,the smooth muscle thickness and the deposition of collagen fibers under airway epithelium were significantly increased(P<0.01).Compared with the model group,the airway inflammatory infiltration in Yanghe Pingchuan group,inhibitor group and dexamethasone group was significantly reduced,and the total thickness of airway wall,the smooth muscle thickness and the deposition of collagen fibers under airway epithelium were significantly decreased(P<0.05).There was no difference among Yanghe Pingchuan group,inhibitor group and dexamethasone group(P>0.05).3.Compared with the normal group,the levels of serum total Ig E,OVA specific Ig E,PDGF and TGF-?1 in the model group increased significantly(P <0.01).Compared with the model group,the levels of serum total Ig E,OVA specific Ig E,PDGF and TGF-?1 in the Yanghe Pingchuan group,inhibitor group and dexamethasone group decreased significantly(P < 0.01).There was no difference between the Yanghe Pingchuan group,the inhibitor group and the dexamethasone group(P>0.05).4.Compared with the normal group,the expression of Axin protein in the lung tissue of the model group was significantly decreased(P < 0.01),while the expression of?-catenin and Cyclin D1 protein was significantly increased(P<0.01).Compared with the model group,the expression of Axin protein in Yanghe Pingchuan group and inhibitor group was significantly increased(P<0.01),while the expression of ?-catenin and Cyclin D1 protein was significantly decreased(P<0.05).There was no difference in the expression of ?-catenin,Axin and Cyclin D1 between the Yanghe Pingchuan group and the inhibitor group(P>0.05).5.Compared with the normal group,the expression levels of Wnt-3a,Wnt-5a,?-catenin and Cyclin D1 m RNA in the lung tissue of the model group were significantly increased(P < 0.01).Compared with the model group,the expression levels of Wnt-3a,Wnt-5a,?-catenin and Cyclin D1 m RNA in Yanghe Pingchuan group and inhibitor group were significantly decreased(P < 0.05).There was no difference between Yanghe Pingchuan group and inhibitor group(P>0.05).Conclusion:1.Yanghe Pingchuan granules can improve the pathological changes such as airway remodeling in lung tissue of rats with chronic asthma.2.Yanghe Pingchuan granules can regulate the levels of serum cytokines PDGF and TGF-?1 in chronic asthma model rats,and interfere with airway remodeling in chronic asthma model rats.3.Yanghe Pingchuan granules can regulate the expression of Wnt/?-catenin signaling pathway related protein and gene level in lung tissue of chronic asthma model rats.4.The intervention effect of Yanghe Pingchuan granules on airway remodeling and cytokines PDGF and TGF-?1 in chronic asthma model rats may be achieved through the regulation of Wnt/?-catenin signal pathway.
