Objective At present,the drainage tube used in the clinic is mostly made of polydimethylsiloxane rubber(PDMS)material.Due to its hydrophobicity,it may lead to postoperative symptomatic epidural hematoma(SPSEH)due to poor drainage after being placed into the postoperative incision.This issue requires us to improve the hydrophilicity and anticoagulation of the drainage tube through modification.The carboxylated chitosan(CS)has good hydrophilicity and biocompatibility.The hydroxyl carthusa yellow pigment A(HYSA)can prolong the coagulation time,resist platelet aggregation,and is non-toxic and water-soluble.This paper mainly studied the surface hydrophilic anticoagulation modification of PDMS,and explored its mechanical properties,surface hydrophilic surface anticoagulation and biocompatibility,hoping to provide new ideas for the combination of traditional Chinese medicine and biological materials and provide theoretical basis for reducing the occurrence of postoperative hematoma in patients with spinal tumors.Methods In this experiment,the traditional PDMS group was defined as the control group,and the PDMS/CS group,CS Coating group and CS/HYSA Coating group were defined as the experimental group.We use material surface multifunction tester(MFT)test the four sets of samples of modulus of elasticity,and calculate the biggest variables,four groups of samples using dynamic thermal mechanical analyzer(DMA)was developed for the determination of the four groups of sample storage modulus and loss modulus we by field emission scanning electron microscope(FE-SEM)of the four groups of sample surface and cross-section morphology,with multifunctional friction and wear tester(FWT)was developed for the determination of the four groups of sample wall surface coefficient of friction(COF)in the water environment,with optical contact Angle measuring device(CAM)test the walls of the four groups of sample surface water contact Angle(WCA)We also the platelet adsorption of four groups ofsamples and protein adsorption test,and through the four groups of the extract of the sample,the influence of the intervention of plasma coagulation test results to compare traditional drainage tube and drainage tube new anticoagulant hemorrhagic differences between cell proliferation and cell activity test we use to compare traditional drainage tube and the improved biocompatibility of the difference between the new drainage tube.The differences between the control group and the three experimental groups in all the above experimental tests were statistically analyzed,and all the results were statistically significant as p<0.05 or p < 0.001.Results(1)The elastic modulus of the four groups decreased with the increase of the maximum shape variable,and the elastic modulus of the PDMS group was slightly higher than that of the other three groups,but the difference was not important.The energy storage modulus and loss modulus of the four groups decreased with the increase of temperature,and the CS Coating group and CS/HYSA Coating group were slightly higher than the PDMS group and PDMS/CS group;(2)The inner surface and cross section of the four groups of samples are flat and smooth;(3)The surface friction and water contact angles of CS Coating group and CS/HYSA Coating group were significantly lower than those of PDMS group and PDMS/CS group,and the difference was statistically significant(p<0.001);(4)The amount of platelet and protein adsorption on the inner surface of CS Coating group and CS/HYSA Coating group was significantly lower than that of PDMS group and PDMS/CS group,and the difference was statistically significant(p<0.001);(5)The APTT,TT and PT time of CS/HYSA Coating group were all longer than those of the other three groups,while the content of Fib was significantly lower than that of the other three groups,and the above differences were statistically significant(p<0.001);(6)There was no significant difference in cell proliferation and cell activity between the four groups;(7)The release of HSYA in the CS coating increases gradually within 1-3 days,reaches a peak on the third day,then gradually declines and ends around the seventh day.Conclusion(1)Our preparation process can successfully coat the inner surface of PDMS drainage tube with CS hydrophilic coating containing HSYA,and the HSYA in the coating can be successfully released;(2)The results of various experiments proved that the new PDMS drainage tube was similar to the traditional PDMS drainage tube in mechanical properties and biocompatibility,but the former was significantly better than the latter in hydrophilicity and anticoagulation;(3)The improved PDMS drainage tube can meet the clinical needs of SESPH patients and is expected to help reduce the occurrence of SESPH in patients with spinal tumors in the future;(4)This experimental study is a feasible attempt to improve the kinetic behavior of HSYA in vivo,and makes a new attempt for the application of traditional Chinese medicine in the field of biomaterials.We hope that this experimental study can provide new ideas and methods for the combination of traditional Chinese medicine and modern medicine.
