Analysis Of Therapeutic Effect Of Arsenic Trioxide Eluting CalliSpheres Microspheres In TACE Treating The Rabbit Liver Tumor Models

PurposeTo study the therapeutic effect of CalliSpheres Beads(CB)loaded Arseni c trioxide(ATO)on rabbit VX2 liver tumor by transcatheter arterial chemoem bolization.Materials and Methods120 rabbits with VX2 liver tumors were randomly divided into 4 groups:CB group,CBATO group,cTACE group and control group.Five rabbits in ea ch group were sacrificed at 12 hours,3 days,7 days and 14 days after operat ion.The normal liver and tumor tissues were taken for pathological,immunohi stochemical and apoptosis and proliferation of tumor cells examination.10 rabbi ts in each group were left for survival observationResults14 days after TACE,the total carcinoma volume(CV)and tumor metastas is rate of CBATO group were significantly lower than those of the other three groups,while the survival time of CBATO group was longer than that of othe r three groups.3,7,14 days after TACE,hepatocyte growth factor-1 ?(HIF-1?),vascular endothelialgrowth factor(VEGF)and micro vessel density(MVD)in residual tumor area of CBATO group was lower than CB and cTACE group.7,14 days after TACE,matrix metalloproteinase 9(MMP-9)in residual tumo r area of CBATO group was lower than that in other three groups.7,14 days after TACE,the apoptotic rate of residual tumor cells in CBATO group was hi gher than that in other three groups,but all days after TACE,the proliferation index(PI)of residual tumor cells in CBATO group was lower than that in oth er three groups.7,14 days after TACE,the expression of E-cadherin in CBATO group was more than CB group and cTACE group,while the expression twist protein,N-cadherin and Vimentim were less.ConclusionTACE using CBATO can significantly improve the necrosis rate of rabbit VX2 liver tumor,inhibit the growth and metastasis of rabbit VX2 liver tumo r,and prolong the survival period of experimental animals.
