Objective:The occurrence of skin photoaging is related to the over-generation of reactive oxygen species(ROS)induced by ultraviolet(UV)and the inflammatory response,therefore,anti-inflammatory and anti-oxidation can be used as a treatment for skin aging methods.As the main active ingredient in ginseng,total saponins of Panaxginseng have a variety of pharmacological effects such as anti-oxidation and anti-aging.However,the poor skin transmissibility of total saponins of Panaxginseng limited its development and utilization.Liposome,a new drug delivery technology,was used to encapsulate total saponins of Panaxginseng in order to improve skin transmissibility and make it work better.By observing the therapeutic effect of total saponins of Panaxginseng on photoaging skin of rats,the possible therapeutic mechanisms were preliminarily discussed from the perspectives of histopathology,oxidative stress and inflammatory response.Methods:Firstly,an in vitro analysis method of total saponins of Panaxginseng was established:The content of total saponins of Panaxginseng was determined by HPLC.A method for the determination of encapsulation was established.Total saponins of Panaxginseng liposomes were prepared by ethanol injection method,and the free drug and carrier liposome of drug were separated by ultrafiltration centrifugation method.The method of single factor investigation was used to optimize the formulation and preparation of total saponins of Panaxginseng liposomes.Quality evaluation and Study on skin permeability in vitro were carried out.Through the establishment of light aging rats model,effect of total saponins of Panaxginseng on photoaging skin of rats preliminarily evaluated from the appearance shape and thickness of skin.Photoaging skin of rats was observed the skin morphology changes after HE and aldehyde fuchsin staining.The content of MDA,gsh-px,SOD enzyme activity,matrix metalloproteinases MMP-1 and MMP-3,inflammatory cytokines IL-1β,IL-6 and tumor necrosis factor TNF-α in the skin tissue of rats were determined.The possible mechanism of total saponins of Panaxginseng treating rats photoaging skin model was preliminarily discussed.Results: The best prescription for total saponins of Panaxginseng liposomes is: the molar ratio of Lecithin Hydrogenated to total saponins of Panaxginseng is 1: 0.4;the molar ratio of cholesterol to Lecithin Hydrogenated is 1: 3.5;the volume ratio of organic phase to water phase is 1: 9;Stirring time: 4h;The average encapsulation rate of total saponins of Panaxginseng liposomes measured by confirmatory experiments: 63.38 %,average particle size is 111.0 nm;PDI: 0.059,Zeta potential-5.72 mv;The encapsulation efficiency of total saponins of Panaxginseng liposomes lyophilized powder was 61.21 %.The results of in vitro transdermal test showed that,total saponins of Panaxginseng solution was no transmission within 24 hours.The total transmission rate of total saponins of Panaxginseng liposomes in skin was 85.46 % within 24 hours.The effect of total saponins of Panaxginseng on photoaging skin in rats was compared with the model group.The skin of the plastid group and the solution group improved in different degrees.The HE and aldehyde fuchsin staining results showed that the total saponins of Panaxginseng could effectively improve the skin cuticle thickness,elastic fiber breakage,and agglomeration caused by UV.Compared with the control group,the content of MDA in the skin tissue of the total saponins of Panaxginseng liposome group decreased significantly,the activities of GSH-Px and SOD enzymes increased significantly,The levels of inflammatory factors IL-6 and IL-1β And tumor necrosis factor TNF-α,and the contents of matrix metalloproteinases MMP-1 and MMP-3 were significantly increased.Conclusion: The preparation technology of saponins of Panaxginseng liposome was optimized by single factor method and its quality was evaluated.The results of in vitro transdermal experiments showed the poor skin penetration of total saponins of Panaxginsenge and liposomes can improve it significantly;topically using total saponins of Panaxginseng liposomes has a certain therapeutic effect on photoaged skin in rats.This protective effect may be related to inhibiting UV-induced skin lipid peroxidation,increasing the activity of antioxidant enzymes SOD and GSH-Px,and reducing the inflammatory factors TNF-α,IL-1βand IL-6 expression,reducing the content of MMPs.
