Study On The Effect Of Yiqi Huoxue Tongluo Formula Based On FXR/FGF15 Signaling Pathway On Glucose And Lipid Metabolism In Type 2 Diabetic Mice

Objective:In this study,FXR / FGF15 was used as the target,starting from the key link of bile acid metabolism pathway,and T2 DM model mice of type 2 diabetes were used as the research object.By using the method of molecular biology,the role and potential molecular mechanism of Yiqi Huoxue Tongluo Recipe in the treatment of type 2 diabetes were revealed,so as to explore the new mechanism of type 2 diabetes and the new strategy of prevention and treatment.Methods:ICR male mice were selected and fed with high-fat and high-sugar diet for 8weeks to establish T2 DM animal model combined with chemical inducer streptozotocin.after modeling successfully,the model mice were randomly divided into high-dose group,low-dose group,model group and metformin group according to random digital table method.in addition,12 healthy mice of the same batch were selected as normal group.The formula of administration is gavage,every 7 days regular test mice weight,blood sugar.mice were sacrificed after 8 weeks of gavage.Changes of blood glucose,blood lipids,bile acid levels and FXR?FGF15 were detected in mice.Results:1.Compared with the model group,mice with high dose group,low dose group and metformin group had significantly improved blood glucose and blood lipid(P<0.05);2.Compared with the normal group,the total bile acid in the treatment group increased significantly(P < 0.05),and the high dose group was more obvious.3.Compared with the model group,the expression of fxrand FGF15 protein in DM mice in the high dose group and the low dose group were increased.Conclusion: Yiqihuoxuetongluo recipe can effectively reduce the blood glucose and blood lipid of T2 DM mice,and its mechanism may be related to the regulation of bile acid signal pathway by Yiqihuoxuetongluo recipe and the up regulation of FXR and FGF15 levels.
