| Purpose:Experiment 一:This experiment starts from the experiment in vitro to explore the effect of Fuzheng Kangai Decoction containing serum on the proliferation and apoptosis of A2780 ovarian cancer cells.Testing β-catenin and c-Myc protein to confirm that Fuzheng Kangai Decoction can inhibit the growth and promot apoptosis of A2780 human ovarian cancer cells,it may be related to the regulation of key proteins in Wnt/β-catenin signaling pathway.Experiment 二 : Through the action of quercetin,the effective component of Fuzheng Kangai Decoction,on A2780 ovarian cancer cells,it is proved that quercetin can inhibit the proliferation and induce the apoptosis of ovarian cancer cells,which will open up a new way for the treatment of ovarian cancer and the development of new drugs.Materials and methods:Experiment 一 : Inhibitory effect of Fuzheng Kangai Decoction containing serum on proliferation and apoptosis of ovarian cancer A2780 cells through Wnt/β-catenin signaling pathway related proteins.1 Preparation of containing serum and cell cultureSPF grade SD rats were randomly divided into two groups,with 10 rats in each group.The experimental group was given Fuzheng Kangai Decoction,which was concentrated 6times,and then gavaged 9 times of the equivalent dose of human.The control group was given physiological saline by gavage and continuous gavage.On the tenth day,1 hour after gavage,blood was taken under anesthesia to prepare containing serum and blank serum for cryopreservation.Ovarian cancer A2780 cells were cultured in an incubator(saturated humidity,37 ℃,5%CO2),and the cell density was observed under a naked eye microscope,quickly spreading the bottom of the culture flask for passage.2 Effect of Fuzheng Kangai Decoction containing serum on proliferation and apoptosis of A2780 cells.The containing serum of Fuzheng Kangai Decoction and the blank serum were mixed according to the ratio of one time dilution from the high concentration group in turn,and marked as high,medium and low concentration groups.CCK-8 was used to detect cell proliferation;Cell apoptosis was detected by flow cytometry;Western blot detection of each group β-catenin and c-Myc protein.Experiment 二:Effects of Quercetin on Proliferation and Apoptosis of Ovarian Cancer A2780CellsOvarian cancer A2780 cells were cultured in vitro and divided into quercetin group and control group.The quercetin group was treated with 0μmol/L,10μmol/L,20μmol/L,40μmol/L,80μmol/L,160μmol/L and 320 μmol/L after 24 h of intervention with the cells,the cell inhibition rate was calculated using the cck-8 method.After quercetin was applied to ovarian cancer cells at 0μmol/L,40μmol/L,80μmol/Land160μmol/L for 24 hours,the apoptosis of ovarian cancer cells was detected by flow cytometry.Results:Experiment 一1 Cell proliferationThe 24 h inhibition rate of serum containing Fuzheng kangai Decoction: the inhibition rate of high concentration group was 24.91%,the inhibition rate of medium concentration group was 19.8%,and the inhibition rate of low concentration group was 15.36%.Compared with the control group,the drug-containing serum group had a significant effect on inhibiting the proliferation of ovarian cancer cells(P<0.05).2 Cell apoptosisThe total apoptosis rate in the high concentration group of Fuzheng kangai Decoction was 6.34%;The total apoptosis rate in the medium concentration group was 4.28%;The total apoptosis rate in the low concentration group was 3.52%.Compared with the control group,the difference was statistically significant(P<0.05).Fuzheng kangai Decoction can promote the apoptosis of ovarian cancer cells.3 β-catenin and c-Myc proteinThe results of Western Blot method show that the expression of β-catenin and c-myc protein in the high concentration group of Fuzheng Kangai Decoction decreased,and the band became narrower,compared with the control group,the difference was statistically significant(P < 0.05),but there was no significant change in the middle and low concentration groups,and the difference was no statistical significant(P > 0.05).Experiment 21 Quercetin inhibits the proliferation of ovarian cancer cellsQuercetin can inhibit the proliferation of A2780 cells,compared with the blank control group,at 20μmol/L,40μmol/L,80μmol/L,160μmol/L,320 μmol/L groups,the inhibition rate of ovarian cancer increased gradually,and there was significant difference between the groups(P<0.01).With the increase of quercetin concentration,the inhibition rate of ovarian cancer increased.2 Effect of quercetin on apoptosis of A2780 cellsWith the increase of quercetin concentration,the apoptosis rate of cells in each group also increased in a dose-dependent manner.Compared with the control group,there was a significant difference in statistics between each group(P<0.01),indicating that quercetin can induce apoptosis of A2780 cells.Conclusion:1.Fuzheng kangai Decoction can inhibit the proliferation of ovarian cancer cells and promote the apoptosis of ovarian cancer cells.2.Fuzheng kangai Decoction may pass Wnt/ β-Catenin signaling pathway inhibits the growth of ovarian cancer cells.3.The effective ingredient quercetin in Fuzheng kangai Decoction can inhibit the proliferation of ovarian cancer cells and promote apoptosis of ovarian cancer cells. |
