| Background and Objective MicroRNAs(miRNAs)are a class of endogenous small nucleic acid molecules,which can mediate neurovascular remodeling and enhance functional recovery after stroke.Up-regulation of microRNA-19b was found in subventricular zone(SVZ)neural progenitor cells after stroke,and the overexpression of microRNA-19b can promote the proliferation and differentiation of neural stem cells.At the same time,miR-19b-enriched exosome treatment had significantly more robust effects on improvement of neurological function and enhancements of neurogenesis,and neurite remodeling/neuronal dendrite plasticity in the ischemic boundary zone(IBZ)than the control MS C exosome treatment.It was suggested that overexpression of the microRNA-19b could promote the recovery of neurological function after stroke.However,the expression of microRNA-19b can undergo different changes over time as well as different from subtypes of stroke.The changes of plasma microRNA-19b expression in patients with ischemic stroke not clear,and whether it is related to the recovery of neurological function have not been reported.This study aims to dynamically observe the changes of plasma microRNA-19b-3p expression in patients with ischemic stroke,and to explore the relationship between microRNA-19b-3p expression and the recovery of neurological function after stroke.Methods Patients within 3 days initial infarction onset and with neurological deficiency manifestation such as speech disorder,limb paralysis and numbness were enrolled as the observation group.Healthy volunteers with gender and age matched the patients were chosen as the control group.All patients underwent plasma microRNA-19b-3p detection at three time points,within three days(d3),fourteen days(d 14),twenty-eight days(d28)after infarct onset,while controls underwent the same plasma microRNA-19b-3p detection just one time.At three time points,all patients were evaluated with National Institutes of Health Stroke Scale(NIHSS),Barthel Index(BI)to assess the changes of neurological function deficits and the ability of daily living by time.The relative expression of plasma microRNA-19b-3p was calculated by 2-△△Ct at three time points.To observe the changes of mir-19b-3p and nerve function score over time in patients with ischemic stroke,RMANOVA was used for analysis.To investigate the relationship between the percent change of microRNA-19b-3p and of nerve function score after 28 days infarct onset,Spearman correlation analysis was used,with a formula of[(d28-d3)/d3]x 100%to calculate the percent change.Results A total of 21 patients with ischemic stroke were collected,including 15 males and 6 females.There was no significant difference in age between two groups.The ratio of hypertension in patients with ischemic stroke was significantly higher than that in the control group.Classification results of TOAST showed that there were 18 cases of large artery atherosclerosis,2 cases of cardioembolism,and 1 case of stroke of other undetermined etiology.There were 14 cases of anterior circulation cerebral infarction and 20 cases of paralysis.During the observation period,the NIHSS score of patients with ischemic stroke decreased with time,while the Barthel index increased with time.The relative expression of plasma microRNA-19b-3p in patients with ischemic stroke was lower than that in the control group during the observation period,and gradually increased with time.Spearman correlation analysis showed that the increased percentage of microRNA-19b-3p negatively correlated with the decreased percentage of NIHSS,but positively correlated with the increased percentage of the percent change of BI scores.Conclusion The present study indicates that the expression of plasma microRNA-19b-3p decreased initially and then increased gradually after stroke.And the increased expression of microRNA-19b-3p may promote the recovery of neurological function. |
