Objective: To study the therapeutic effect of compound glycyrrhizin(commercial name: Meineng)combined with swimming exercise on non-alcoholic fatty liver disease(NAFLD)rat model.Methods: Forty clean SD rats aged 6-8 weeks were selected.After 1 week of adaptive feeding,they were randomly divided into 5 groups,8 in the control group(CON)were fed in ordinary during 12 weeks;32 in the high-fat group,after 12 weeks of fat-feeding,8 rats in the model group(HC),8 rats in the glycyrrhizin group(HG),8 rats in the swimming exercise group(HE),and 8 in the groups of glycyrrhizin and swimming exercise(HGE).Two rats were randomly selected from the CON,HC,HG,HE,and HGE groups separately,and their livers were removed for pathological analysis to determine the success of modeling,and all remaining rats were weighed.After the successful modeling,a formal experiment was performed for 8 weeks.The rats in the CON group continued to be fed with ordinary feed;the rats in the HC group were fed with a high-fat diet;Rats in the HE group were given 90 minutes of swimming exercise every afternoon;rats in the HGE group were given 3ml of compound glycyrrhizin while being given 90 minutes of swimming exercise every afternoon.During the experiment of each group of rats,the CON group ate the ordinary diet,the other groups continued to eat the high-fat diet,and each group was free to drink purified water except for the time of gastric administration.After the last intervention,rats in each group were fasted for 24 hours,sacrificed after anesthetized with chloral hydrate,and tested for serum Alanine aminotransferase(ALT),Aspartate aminotransferase(AST),and Gamma-glutamyltransferase(?-GGT),Tumor necrosis factor(TNF-?),Iterleukin-6(IL-6),Hyaluronic acid(HA),Laminin(LN),the contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in the liver tissue grinding fluid were measured,and liver pathological sections were made to detect fatty liver disease.Results: The 12-week high-fat diet could replicate the NAFLD rat model.Compound glycyrrhizin gavage and swimming exercise had therapeutic effects on NAFLD rats.In terms of serological indicators,the ALT,AST,and ?-GGT in the HE and HGE groups were significantly lower than those in the HC group.The HGE group was lower than the HG and HE groups.(P <0.001).Among the HA and LN indicators,the HC group was significantly higher than the other groups,the difference was statistically significant(P <0.001),but the HGE group was not statistically significant compared with the HG and HE groups.The serum inflammation indicators TNF-? and IL-6 in the HG,HE and HGE groups were lower than those in the HC group.The HGE group was the lowest among the three groups,but all were higher than the CON group,the difference was statistically significant(P <0.001).There was no difference between the HE and HG groups.In terms of body weight,the rats after modeling were significantly higher than those in the CON group,the HGE and HE groups were lower than the other groups,and the rats in the HG group were higher than the swimming intervention group,and the differences were statistically significant(P <0.001).The liver and pathology of the rats in the treatment group were better than those in the HC group.The liver index and NAS score in the HC group were significantly higher than those in the other groups,but there was no statistical significance between the HGE,HE and HG groups.In the SOD test among liver tissue homogenates,the CON group was the highest,followed by the HGE group,the HE group,and the HG group,and the HC group was the lowest.The differences between the CON group and the HC group,and the HC group with the HG,HE,and HGE groups were statistically significant.(P <0.001).In the MDA test of liver tissue homogenate,the HC group was significantly higher than the other groups,the HGE group was lower than the HG and HE groups,and the difference was statistically significant(P <0.001).The difference between the HE and HG groups was not significant.Conclusion: Compound glycyrrhizin and swimming exercise can improve liver function,reduce serum inflammation indexes,enhance liver antioxidant capacity,improve liver index,reduce liver wet weight,and improve liver pathological conditions to a certain extent.In terms of improving liver function,reducing serum inflammation indicators,and enhancing liver antioxidant capacity,compound glycyrrhizin combined with swimming exercise has a better therapeutic effect on NALFD rats than on its own.
