Objective: To investigate the expression of Indian hedgehog(Ihh)in the serum and synovial fluid of Knee Osteoarthritis(KOA)patients,to understand the correlation between Ihh and the duration,diagnosis and classification of KOA patients,and to explore its clinical application value.Methods: Patients with KOA were selected as subjects and subjects with normal knee joints as controls.Inclusion and exclusion criteria were set according to the purpose of this study and the KOA-related medical guidelines.KOA patients treated in the outpatient and inpatient departments of our hospital were collected.A total of 243 KOA patients treated and treated in the outpatient and inpatient departments of our hospital were collected in accordance with the sodium exclusion standard.115 serum samples and 167 samples of joint fluid were collected from the experimental group,among which 39 serum samples and 39 samples of joint fluid were taken from the same patients.The subjects' knee X-ray was read and K&L grading was performed.The patients were divided into acute,subacute and chronic groups according to the course of disease.Serum samples from 103 patients with normal knee joints were collected in accordance with the naturopathy standard and set as the control group(the control group had no samples of joint fluid).The contents of Ihh in the samples were determined by enzyme-linked immunoassay(ELISA)and statistically analyzed by SPSS26.0 software.Results:(1)The serum Ihh concentration in KOA patients was significantly higher than that in patients with normal knee joint,and the difference was significant(P < 0.0001).(2)The Ihh concentration in serum and joint fluid of the same KOA patients tended to be consistent,with no statistical difference was found between the two groups(P > 0.05).Serum and Ihh concentrations in joint fluid of patients with the same grade of K&L also tended to be consistent,with no statistical difference was found between the two groups(P > 0.05).(3)The serum Ihh content in KOA patients was not related to the severity of the disease,and there was no correlation between Ihh concentration and K&L level,and there was no significant difference between each group(P > 0.05).The Ihh content in synovial fluid was also independent of the K&L grade.(4)The serum Ihh concentration was independent of the duration of the disease,and there was no significant difference in serum Ihh concentration between the acute,subacute and chronic groups(P > 0.05).Ihh concentration in joint fluid was also independent of duration.Conclution: The Ihh concentration of serum and synovial fluid can be used as a laboratory reference for the diagnosis of KOA.The Ihh concentration in serum and joint fluid of KOA patients was basically the same,and was independent of the course and severity of the disease.
