Objective:Detect normal rats liver metabolites and non-alcoholic fatty rats liver me tabolites by using Ultra High Performance Liquid Tandem Chromatography Quadrupole Time of Flight Mass Spectrometry,observe the effect of Shenling Baizhu powder on non-alcoholic fatty liver metabolite look for different metabolites and related metabolic pathways,explore the mechanism of Shenling Baizhu Powder in treating non-alcoholic fatty liver rats.Methods:96 male SPF SD rats were grouped and administered by gavage: normal group and model group are give normal saline,feninuobeite group(18mg/kg),Shen ling Baizhu powder low dose group(1.8g/kg),Shenling Baizhusan medium dose group(3.6g/kg),Shenling Baizhusan high dose group(7.2g/kg)are give the corresponding drugs.Feeding method: In addition to the normal group feeding with normal feed,the other groups are fed with high-fat feed.At the 8th week and 12 th week after gavage,samples are take to detect serum total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),Alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),blood glucose(Glu)content;liver pathological sections were made,HE staining and oil red Ostaining.Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UHPLC-QTOFMS)is used to detect endogenous small molecules in the liver of each group of rats,combined with principal component analysis(PCA)and partial least squares(OPLS-DA)and other methods for pattern recognition of meta bolite profile data,analysis and identification of metabolic markers,through enrich ment analysis and topological analysis and other methods to screen from the KEGG database to find the key metabolic pathways with the strongest correlation with metabolite differences.Results:The liver body index of rats in the normal group increased gradually,the liver body index of the model group increased the most.Each drug group was bet ween the normal group and the model group.Liver pathological results suggest that there is no obvious abnormality of hepatic cord arrangement and hepatic lobular structure in each dose group of Shenling Baizhu powder,and the degree of hepatic cell fatty degeneration and the amount of fat particles are significantly reduced.Serum biochemical results suggest that: compared with the normal group,the model g roup TC,TG,LDL-C,ALT,AST,TBIL are higher than the normal group,the model group HDL-C is lower than the normal group,P<0.05 difference is statistically significant.Compared with the model group,the TC,TG,LDL-C,ALT,AST,and TBIL of the fenofibrate group and Shenlingbaizhu powder were lower than the model group,the fenofibrate group and Shenling Baizhu powder were all lower than the model group.The HDL-C of the group was higher than that of the model grou p,the difference of P<0.05 was statistically significant.Liver metabolomics results: There were 26 different metabolites in the liver tissues of the model group and the normal group compared to the liver metabolites;there were 32 different metabolites in the Shenling Baizhu powder group and the model group.Compared with the normal group,the main different metabolites of the model group can be divided into the following categories: organic acids,amino acids,lipids,nucleotides,amines;the comparison between the Shenling Baizhu powder group and the model group mainly includes: organic acids,nucleosides Acids,amino acids,lipids,sugars,purines,vitamins.The above differential metabolites were compared through databases,metabolic pathway analysis and literature search,foundth at the metabolites with obvious multiples of differences were: pyruvic acid,L-palmitoyl,sterol,maltotriose,stachyose,arginine succinicacid.Among them,the model group was compared with the normal group: pyruvate,L-palmitoyl,and de-sterol contents were increased;the Shenling Baizhu powder group was compared with the model group: pyruvate,L-palmitoyl,and de-sterol contents were reduced.The above differential metabolites may become candidate biomarkers for the diagnosis and treatment of non-alcoholic fatty liver disease.These different metabolites are strongly related to the following three metabolic pathways: glycerophospholipid metabolism,pentose phosphate metabolism,and glycine,serine,and threonine metabolism.Thes e pathways are closely related to lipid transport and metabolic transformation.Conclusion:1.Replicated the high-fat diet non-alcoholic fatty liver rat model Succe ssfully.2.Shenling Baizhu powder can reduce liver cell lipid deposition,reduce blood lipids,improve liver function in non-alcoholic fatty liver rats,and play a therapeutic role in non-alcoholic fatty liver.3.Shenling Baizhu Powder can affect the metabolism of sugars,lipids and amino a cids,enhance the body’s transport and metabolic transformation of lipids,promote the transport of fine substances in the spleen of traditional Chinese medicine,and thus play a preventive and therapeutic role in non-alcoholic fatty liver.
