Objective:This experiment chooses Safflower,persicae,achyranthes bidentata and other activating blood circulation herbs widely used in the treatment of hepatocellular carcinoma in clinic,Choosing anti-angiogenesis effect herbs in TG flila-EGFP transgenic zebrafish model.It systematically and impersonally estimated the reliability and security of Safflower,persicae, achyranthes bidentata and other activating blood circulation herbs used in the treatment of hepatocellular carcinoma. The test provided a reliable basis for curing hepatocellular carcinoma with activating blood circulation herbs.Methods:Chossing TG flila-EGFP transgenic zebrafish model as a biological research model, Each sample with 30 zebrafish embryos in 12 orifice (Falcon) in the experiment. The concentration of 200 ?g/ml aqueous extract of Safflower,persicae, achyranthes bidentata and positive control drug PTK787 (5?g/ml) respectively dealing with 22h after post-fertilization (hpf) embryos until 48h. Completely link between dorsalis aorta (DA) and back long axis vessels (DLAV) is the interconnected somite vessels (ISV), which is an important indicators of zebrafish angiogenesis (angiogenesis). vascular phenotype were observed and photographed after medication by a stereoscopic microscope (bright field) and stereo fluorescence microscope.after 48hpf photograph, analyzing interconnected somite vessels effected by each sample.meanwhile, observing and counting the developmental toxicity and mortality of Zebrafis induced by each sample.Results:No developmental toxicity or death in each sample induced by drugs.whole ISVs phenotype were observed by a stereoscopic microscope (bright field) and stereo fluorescence microscope:Blank group of zebrafish embryo ISVs fluorescence expression is complete and clear. Positive Vatalanib group of zebrafish embryo lose ISV and DLAV fluorescence expression compared with blank group; The aqueous extract of Safflower,persicae,and achyranthes bidentata group of zebrafish embryo ISVs and DLAV fluorescence expression is different degrees of thinner, fluorescence intensity is abate, and distribution of the disorder.one of the aqueous extract of Safflower, achyranthes bidentata group of zebrafish embryo ISVs fluorescence expression is thinner,obvious strength reduction,and distribution of the disorder.Especially the aqueous extract of achyranthes bidentata group.Conclusion:Vatalanib has anti-angiogenesis effect in the ISVs and DLAV of transgenic zebrafish embryo model; Safflower,persicae,and achyranthes bidentata have different degrees of inhibiting effect. Safflower, achyranthes bidentata are more obvious,especially achyranthes bidentata,and safely and reliably,no teratogenesis or death;Treatment for hepatocellular carcinoma is safe and reliable by using Safflower,persicae,achyranthes bidentata and other activating blood circulation herbs in clinical.
