Ganoderma lingzhi has been used as a traditional Chinese medicine,which shows various medicinal effects such as antitumor,antiviral,anti-inflammatory,antioxidant,hepatoprotective,anti-diabetic activities.Polysaccharides and triterpenoids are the main active components of Ganoderma lingzhi.The content of triterpenoids is one of the important indicators for the quality of Ganoderma lingzhi.In this thesis work,the separation,purification,qualitative and quantitative analysis of Ganoderma triterpenoids are studied,and the main results are summaried as follows:1.The contents of polysaccharides and triterpenoids in different parts(whole fruiting body,pileus,stipe,context,tube and spores)of Ganoderma lingzhi(Jinzhai)are evaluated.Determined by sulfuric acid-anthrone method,the polysaccharides content in the sporoderm-broken spores is the highest,and the polysaccharides content in stipe is higher than that in pileus.For evaluation of triterpenoids based on the HPLC measurement,the content in the pileus is higher than that in the stipe,and the content of triterpenoids in the context is higher than that in the tube.In particular,the content of ganoderic acid A(GAA)is different in different parts of the fruiting body,and the content of GAA is highest in pileus.2.The method of determining the Ganoderma triterpenoids by UV-Vis absorption spectroscopy based on the colorimetric reaction between triterpenoids with perchloric acid and vanillin is revisited and revised.The standard curves for the triterpenoids are measured and optimized.Several main triterpenoids purified from Ganoderma lingzhi fruiting body are analyzed,and our results indicate that if coleanolic acid is used as standard to evaluate the triterpenoids,the evaluation results are much less than the true values.We also obtain the better linear relationship of the standard curve based on integrating the area of the UV-visible absorption bands.3.Ganoderma triterpenoids are isolated and purified from Ganoderma lingzhi by column chromatography,and several triterpenoids mixtures are examined.Analysis of the purified triterpenoids using middle-infrared(mid-IR)and near-infrared(NIR)spectroscopy indicate that the characteristic peaks of mid-IR spectroscopy are mainly in the regions of 2960-2850 cm-1,1500-1800 cm-1 and 1500-1200 cm-1,while the NIR characteristic peaks mainly include the 5920 cm-1,5780 cm-1,4415 cm-1,4340 cm-1 and 4254 cm-1 bands.Besides,based on the GAA content of fruiting body determined by HPLC and the spectral features of NIR spectra,the possibility of NIR quantification of ganoderic acids in Ganoderma lingzhi is proposed.
