Chinese medicine is a national treasure of my country,and its green source is safe and efficient.At present,the adverse reactions of synthetic drugs limit their clinical application,and the development of chemotherapeutic drugs has a slow and difficult cycle,so research on traditional Chinese medicine is in full swing.Modern nano drug delivery systems tend to mature and have many advantages.Nano drug delivery systems based on traditional Chinese medicine have attracted widespread attention.Colchicine is a plant alkaloid with a long history.It was first used to treat acute attacks of gout and rheumatism.However,due to the narrow clinical treatment index,it is necessary to take more drugs,which leads to patients who need to be cautious and experience poorly.More importantly,taking colchicine causes serious side effects in the gastrointestinal tract.Once colchicine poisoning has serious consequences and there is no rescue agent.As the research found that colchicine is an effective anti-tumor microtubule-targeted drug,the research and application of colchicine in tumor therapy was gradually carried out.In this paper,colchicine is taken as the main research subject,and the nano-drug delivery system based on colchicine is explored and constructed,and the preparation scheme is optimized for characterization,and finally the anti-tumor effect is investigated.1.Preparation and Characterization of Polycarbonate of ColchicineThe polycarbon spots of colchicine were prepared by hydrothermal method.The effects of hydrothermal temperature,reaction time and the amount of colchicine added on the fluorescence performance of polycarbon spots were investigated to determine the best preparation conditions.The measurement of the laser nanometer particle size analyzer showed that the prepared carbon dots had a particle size of about 160 nm,and the morphology results showed spherical particles with uniform dispersion.The fluorescence spectrophotometer has a fluorescence dependence and can emit fluorescence under 365 nm ultraviolet irradiation,which can be seen as bright blue-green fluorescence by the naked eye.After surface structure analysis,it can be speculated that the colchicine polycarbon dot has a hydrophilic group and exhibits excellent water solubility.Cell results show that it has excellent biocompatibility and can be used as a safe and nontoxic carrier.The colchicine polycarbon dots are used as nanocarrier materials to load the anticancer drug doxorubicin,and they are mainly combined by electrostatic adsorption.The particle size and PDI of the system after drug loading are both small,and the drug loading is about 3.2%.Cell experiments have shown that the preparation of colchicine as a polycarbonate can reduce the toxicity of the original drug colchicine on human breast normal epithelial cells,and the formation of colchicine polycarbohydrate-loaded doxorubicin system can inhibit MDA-MB-231 cell vigor,with certain anti-breast cancer activity.2.Preparation and characterization of chitosan-colchicine microspheresChitosan-colchicine nanospheres were successfully prepared by ion cross-linking method.The drug loading capacity,encapsulation rate,yield and particle size were used as the screening conditions for single-factor investigation.Together with the results of orthogonal experiment,the microspheres The optimal preparation conditions are chitosan concentration 2mg/ml,sodium tripolyphosphate concentration 1.5mg/ml,and chitosan to colchicine feeding ratio of 4:1.According to the analysis of particle size and morphology,the prepared nano-microspheres have a particle size of about 150 nm,a small PDI value,and are uniformly dispersed spherical particles with a drug loading of about 13.8%.Drug release experiments were carried out on the drug-loaded microspheres,and the results showed the characteristics of slow release without obvious burst release.The CCK-8 experiment showed that the chitosan-colchicine nanospheres have better biocompatibility with the naked drug colchicine on bone cells,but it has certain toxicity to MDA-MB-231 cells and can inhibit breast cancer cells.Growth.
