Objective:To investigate the effect of Aconitum injection on the expression of BMP-2 and GSK-3? in articular cartilage of knee osteoarthriitis(KOA)model rabbits,and explore the mechanism of Aconitum injection in the treatment of KOA and provide theoretical basis for clinical application of Aconitum injection in the treatment of KOA.Methods:Thirty rabbits were randomly divided into three groups:blank group.model group.sodium hyaluronate group,Zhengqinafengtongning group and Aconitum injection group?The rabbits in the model group,sodium hyaluronate group,Zhengqingfengtongning group and Wutou injection group were injected with 2%papain solution-0.03 respectively on the 1st,4th and 7th day,except the blank group The koa model was established by L-cysteine solution(2:1)of mol/L.and 0.1ml kg of saline,sodium hyaluronate,Zhengqingfengtongning and Aconitum injection were injected into the articular cavity on the 1st.4th and 7th day after the model was established?At the end of administration,the above-mentioned five groups of rabbits were respectively extracted joint fluid to observe the morphology of the knee joint cartilage with naked eves;the cartilage tissue of the knee joint was taken and stained with hematoxylin eosin method,then the morphological changes were observed under light microscope and Mankin score was made;the immunohistochemistry samples of the soft bone tissue of the knee joint were made with PV two-step method,and the relative expression of BMP-2 and GSK-3? were detectedResults:Compared with the blank group,the edge of the knee joint cartilage in the model group was damaged,and the cartilage surface was damaged;the pathological observation results showed that the joint tissue structure was obviously irregular,chondrocytes were obviously enlarged,there were white bubble like changes,the distribution of cells was disorder?The content of GSK-3? protein in knee joint fluid increased significantly,and the relative expression of BMP-2 in knee joint decreased significantly(P<0.05)?Compared with the model group,the cartilage surface of Aconitum injection group,sodium hyaluronate group and Zhengqingfengtongning group was smooth,and the tissue structure was regular;the content of BMP-2 protein in cartilage was significantly increased,and the relative expression of GSK-3? was significantly decreased(P<0.05),but there was no significant difference between the two groups(P>0.05)Conclusion:1.Aconitum injection has obvious repairing effect on cartilage,which can inhibit the inflammatory reaction in the joint,weaken or even block the degradation of KOA cartilage,reduce the wear of the joint,and delay the development of KOA.2.The mechanism of Aconitum injection may be related to the inhibition of GSK-3? expression and the promotion of BMP2 re lease.3.Aconitum injection can inhibit the synovitis reaction.increase the lubricity of articular cavity.enhance the anti stress level of chondrocytes and inhibit the apoptosis of chondrocytes.
