Objective:By articular cavity injection of aconitum injection for the treatment of rabbit knee osteoarthritis experiment,the comparison between groups of rabbit knee joint cartilage cells,ADAMTS-4,ADAMTS-5 indicators such as expression.To observe the effect of aconitum injection on ADAMTS-4 and ADAMTS-5 in osteoarthritis of rabbit knee joint,explore aconite injection with curative effect mechanism of rabbit knee joint osteoarthritis.Method:1?Papain solution and 2% w/v 0.03 mol/L L-cysteine solution mixture mixed solution in a ratio of 2:1,according to 0.1 ml/kg of the specifications of the injection to the New Zealand white rabbit posterior knee joint cavity,establish the rabbit model of knee osteoarthritis.2 ? 43 male New Zealand white rabbits were purchased and 3 were used for pre-experiment.The remaining 40 were randomly divided into blank group,model group,control group and experimental group.In addition to the blank group,the other groups were made.After the success of the model,the experimental group injected 0.1ml/kg of aconite injection,and the control group was injected 0.1ml/kg with sodium hyalurate injection,and the model group injected 0.1ml/kg of physiological saline,and the blank group did not give any treatment.At the end of the treatment phase,after 1 week,anatomical knee joint was observed.The right posterior articular cartilage was examined by HE staining microscope.The expression of ADAMTS-4 and ADAMTS-5 in articular cartilage was detected by immunohistochemistry.3?Using statistical SPSS19.0 processing analysis software for data processing,measuring data to x ±s said,the comparison between the groups using single factor analysis of variance(one-way ANOVA),P < 0.05 that has a statistical significance.Results:1?Visual observation of cartilage results: Compared with the blank group,the model group found that there were many cracks or defects on the cartilage surface,The edge of the joint almost all the formation of osteophytes,and was yellow;Positive drug control group and experimental group were similar,Compared with the model group,the cartilage surfacecrack or defect was found to be less,Little osteophyte formation,and was pale yellow.2?Microscopic observation Mankin score results: The scores of the model group were higher than those of the blank group(P<0.05),the difference was statistically significant.The positive control group and the experimental group were lower than the model group(P<0.05),The difference was statistically significant.3?The content of ADAMTS-4 and ADAMTS-5 in the articular chondrocytes of the model group was significantly increased with that of the blank group(P <0.05),and the difference was statistically significant.The expression of ADAMTS-4 and ADAMTS-5 in the control group and the experimental group were lower than that in the model group,and the experimental group decreased significantly(P<0.05),and the difference was statistically significant.Conclusion:By lowering the expression of ADAMTS-4,ADAMTS-5 in the articular cartilage of osteoarthritis of the rabbit knee joint,aconite injection plays a role in protecting the cartilage of the knee joint of the rabbit,thereby achieving the goal of treating KOA.
