Objective: The incidence of pulmonary fibrosis is increasing day by day.Modern medicine has no specific drugs for the treatment of pulmonary fibrosis,while traditional Chinese medicine has unique advantages for the treatment of pulmonary fibrosis.Recent studies have shown that some growth factors,such as vascular endothelial growth factor,activate eNOS to release nitric oxide through PI3K/Akt signaling pathway,thus protecting vascular endothelium,maintaining vascular homeostasis and inhibiting pulmonary fibrosis.In this experiment,the effects of Danbei Yifei Decoction on food intake,lung coefficient and eNOS index in rats with pulmonary fibrosis were observed,and the therapeutic effect and mechanism of Danbei Yifei Decoction on rats with pulmonary fibrosis were discussed.Methods: Seventy-two clean male SD rats were randomly divided into six groups,with 12 rats in each group: model control group(Group D),methylprednisolone positive drug control group(Group J),normal control group(Group K),high-dose Danbei Yifei Decoction group(Group A),middle-dose Danbei Yifei Decoction group(Group B)and low-dose Danbei Yifei Decoction group(Group C).Bleomycin atomization inhalation repetitive stimulation model was used.After successful replication of the model,the rats were given routine intragastric administration once a day the next day.Normal control group and model control group were given normal saline.In the high,middle and low dose groups of Danbeiyifei Decoction,the stomach volume of Danbeiyifei Decoction was converted according to the weight of rats according to the ratio of 1.52g/100 g,0.76g/100 g and 0.38g/100 g respectively.Methylprednisolone positive control group was given methylprednisolone solution at a weight ratio of 0.42 mg/100 g.The food intake was recorded every three days,and the food intake of each group was counted.After 14 days of continuous intragastric administration,rats in each group collected 3-4 ml of blood by collecting blood from posterior orbital venous plexus.Samples were taken 28 days after administration.Blood was collected from abdominal aorta.After blood collection,rats were killed by dislocation of cervical vertebrae.The lung tissue was taken out.The lung coefficient was calculated by weighing with analytical balance.The left and right lungs were separated.The right lung was fixed in a jar filled with 4% paraformaldehyde for 24 hours.The tissue was embedded and sectioned.After routine dehydration,tissue transparency,wax immersion,embedding and sectioning,HE and Masson staining were performed.Fibrosis degree and staining results were observed under microscope.Enzyme-linked immunosorbent assay(ELISA)was used to determine rat endothelial nitric oxide synthase.The standard was diluted,added with samples,watered,prepared with liquid,washed,added with enzyme-labeled reagent,washed with water again,developed color,stopped and calculated the actual concentration of eNOS after measurement.The experimental data were statistically analyzed with SPSS statistical software 17.0.Single factor analysis of variance was used in multi-group experiments.Paired sample t test was used for comparison between the two groups.The test level ?=0.05,and the difference was statistically significant when p<0.05.Results: On the 14 th and 28 th day,the serum eNOS of each group was analyzed by ANOVA,the P value was less than 0.01,which proved that the difference of serum endothelial nitric oxide synthase in each group was statistically significant.Paired t test was used to compare the serum eNOS of each group.the results showed that there was significant difference between model control group,danbeiyifei decoction low dose group,methylprednisolone positive control group and normal control group on the 14 th day(p < 0.01).On the 28 th day,the difference between methylprednisolone positive control group,model control group,high,medium and low dose groups of danbeiyifei decoction and normal control group was statistically significant(p<0.01).Compared with methylprednisolone group and model control group,the lung coefficient of each group of danbeiyifei decoction is smaller,but larger than that of the normal control group,which proves that the lung fibrosis of each group of danbeiyifei decoction is recovered and the effect is better than that of methylprednisolone group.The intake of rats with pulmonary fibrosis decreased,and all groups recovered after medication,but the intake of methylprednisolone group was significantly lower than that of danbeiyifei decoction group.Conclusion: 1.Danbeiyifei Decoction can improve the expression level of serum eNOS in rats with pulmonary fibrosis.2.Danbeiyifei Decoction can reduce lung coefficient and improve pulmonary fibrosis in rats with pulmonary fibrosis.3.Pathological observation: Danbeiyifei Decoction can improve the proliferation of fibrous tissues in rats and delay the degree of pulmonary fibers.4.Methylprednisolone may affect the appetite of rats,while Danbeiyifei Decoction can restore the appetite and increase the food intake of lung fiber rats to a certain extent,among which the high dose group has the most obvious effect.
