Cortex Mori(Sang-Bai-Pi)is the dried root bark of Morus alba L..It has been used as traditional Chinese medicine for treating lung heat,asthma,cough,edema,oliguria,etc.In this paper,the main active ingredients of Cortex Mori were refined,and other chemical components are qualitatively studied,while the ethanol extraction process of Cortex Mori was optimized.In addition,the quality evaluation system including thin layer identification,water content,total ash,acid insoluble ash,extract and content determination for Sang-Bai-Pi was established.1.Four main components were prepared from the extracts of Cortex Mori by chromatographic columns with different fillers(macroporous resin,silica gel,MCI,Sephadex LH-20,Sephadex LH-60)and preparative HPLC,and other compounds were separated at the same time.The structures of the compounds were identified by 1H-NMR,13C-NMR and ESIMS.14 compounds were isolated and identified as follows:mulberroside A(1),morusin(2),kuwanon G(3),kuwanon H(4),oxyresveratrol-2-O-β-D-glucopyranoside(5),oxyresveratrol-3′-O-β-D-glucopyranoside(6),xeroboside(7),apiosylskimmin(8),cis-mulberroside A(9),austraone A(10),albanol B(11),mulberrofuran G(12),mulberrofuran B(13),kuwanon J(14).The purity of the four main ingredients(compounds 1 to 4)are all above 95%.2.The fat-soluble components were extracted by ultrasonic method and methyl esterified,the volatile components were extracted by steam distillation method,and then identified them by gas chromatography-mass spectrometry technology and retention indices.39 and 52 constituents from the fat-soluble components and volatile components were identified respectively.The constituents with higher content in fat-soluble components were:α-amyrin acetateandβ-amyrin acetate.The constitutes with higher content in volatile components were D-ellobiose octaacetate.3.The HPLC method was developed for the simultaneous determination of kuwanon G,kuwanon H,and morusin in Cortex Mori.The simultaneous extraction technology of the three components was optimized by response surface methodology.The optimum process parameters for simultaneous extraction of kuwanon G,kuwanon H,and morusin from Cortex Mori were 68%ethanol,14:1 ratio of liquid to material(m L:g),and extraction for 110 min at 74℃.The extraction contents of kuwanon G,kuwanon H,and morusin were 7.09 mg?g-1,1.74 mg?g-1,1.69 mg?g-1,respectively.4.A TLC identification method based on mulberroside A,kuwanon G and morusin of Cortex Mori was established,the method has good specificity.The content of moisture,total ash,acid insoluble ash and extract in Cortex Mori from different origins were determined according to the methods of Chinese Pharmacopoeia of 2015 edition.The final recommendation is that the quality standard of Cortex Mori medicinal materials is:the moisture was less than 13%,the total ash within 9%,acid insoluble ash was not more than 2.5%,the water soluble extract obtained by hot dip method shall not be less than25%.5.A HPLC method was established for the determination of mulberroside A in Cortex Mori.The content of mulberroside A in 85 batches of Cortex Mori was determined.Finally,it is recommended that the content of mulberroside A in Cortex Mori should not be less than 0.91%(On a dry basis).it is suggested that mulberry root with diameter more than 10 mm should be selected as the material part of cortex mori.In addition,it is suggested to select mulberry root with diameter greater than 10 mm as the source of Cortex Mori.The results enrich the research scope of the chemical constituents of Cortex Mori,provide a reference for the efficient utilization of Cortex Mori,and also provide a basis for the quality control of Cortex Mori,which can promote the regulation of the production and utilization of Cortex Mori.
