| Objective:To combine tannic acid(TA)with natural anti-inflammatory effect and ray-crosslinked methacrylic anhydride gelatin hydrogel(GelMA)electrospun fibers to prepare an artificial agent with the function of regulating oxidative stress and suppressing inflammation.Periosteum and its ability to promote bone regeneration in the skull of animal models.Methods:GelMA hydrogel electrospun fibers were prepared by electrostatic electrospinning technology,and then the fibers were self-crosslinked using ultraviolet light cross-linking technology to form a stable fiber membrane structure.The fiber composition structure of artificial periosteum was observed in scanning electron microscope,and GelMA.Fiber and GelMA-TA fiber were characterized by infrared spectroscopy and mechanical instruments.The artificial periosteum was divided into GellMA group,GelMA-TA group and Control group,and co-cultured with rat mesenchymal stem cells(BMSCs)in an incubator.Then,the biological phase of artificial periosteum was measured by CCK8 method,live-staining fluorescent staining Capacitive;use alkaline phosphatase(ALP)staining method,alizarin red staining method,osteocalcin(OCN)immunofluorescence staining method to evaluate the effect of artificial periosteum on the osteogenic differentiation of BMSCs;polymerase chain reaction Technology(PCR)and immunofluorescence staining technique for semi-quantitative analysis to evaluate the anti-inflammatory and anti-oxidative capacity of artificial periosteum.Finally,rat skull defect model animal experiments were performed,and Micro-CT,HE staining,MASSON staining and immunohistochemical staining were used.In this aspect,the anti-inflammatory ability of functionalized artificial periosteum in animals and its effect on osteogenic differentiation were evaluated.Results:GelMA-TA artificial periosteum has better mechanical properties and biocompatibility than pure GelMA fibrous membranes;GelMA-TA artificial periosteum has a stronger ability to promote bone differentiation in BMSCs in vitro cell experiments;In vitro cell experiments showed excellent anti-inflammatory and anti-oxidant capacity;In vivo study,the BV/TV and BMD of the GelMA-TA were higher than those of the other two groups,showing the best ability of bone regeneration,and showed an inhibitory effect on chronic inflammation.Conclusion:Functional artificial periosteal has better mechanical properties and good biocompatibility.In cell experiments,it showed obvious anti-inflammatory ability and ROS clearance ability,promoted the differentiation of BMSCs into osteoblasts and formed stable calcium nodules.In animal experiments,it can promote the differentiation of skull defects into bone repair,new bone activity is more active,and chronic inflammation is suppressed. |
