Objectives:To investigate the therapeutic effect of Corilagin on experimental atherosclerotic minipigs and to explore the regulation effect of Corilagin on protein and mRNA expressions of MMP-1,MMP-2 and MMP-9 in AS plaques,in order to provide more scientific bases and experimental evidences for curing AS with Corilagin.Methods:1.The AS model was established by feeding minipigs with high fatty diet combined with balloon injury.2.Pathological examination was used to observe the effects of different doses of Corilagin on the degree of carotid atherosclerosis in minipigs.3.The expressions of MMP-1,MMP-2 and MMP-9 protein in atherosclerotic plaques of minipigs was detected by immunohistochemistry and Western blot.4.Real-time quantitative polymerase chain reaction(RT-qPCR)was used to determine the effects of Corilagin on the expressions of MMP-1,MMP-2 and MMP-9 mRNA in atheromatous plaque of minipigs.Results:1.The pathological results showed that the vascular intima of AS model group was significantly thickened,a large amount of collagen fibers deposited,endothelial cells were damaged and detached,and there were varying degrees of plaque and foam cells formation on the arterial wall,with lipid deposition.The 2 and 4 mg/kg Corilagin groups can alleviate carotid artery injury and lesions compared with the AS model group.2.The results of immunohistochemistry showed that the protein expressions of MMP-1,MMP-2 and MMP-9 was increased in model group compared with sham operation group.The expressions of MMP-1,MMP-2 and MMP-9 was decreased in as model group after 1,2 and 4 mg/kg Corilagin treatment compared with model group.3.The results of Western blot showed that the protein expressions of MMP-1,MMP-2 and MMP-9 significantly down regulated in atherosclerotic plaque of minipigs by different doses of Corilagin.4.The results of RT-qPCR showed that the mRNA expressions of MMP-1,MMP-2 and MMP-9 down regulated by different doses of Corilagin.Conclusions:1.Corilagin had observe the effect of against the experimental atherosclerosis and showed reduce the degree of arterial vascular injury and lesions,and has a dose-effect relationship.2.Corilagin can significantly down-regulate the protein and mRNA expression of MMP-1,MMP-2 and MMP-9 in atherosclerotic plaque of minipigs.
