Objective:To prepare the TGF-?1 concentration gradient hydrogel scaffold material by simulating the relevant structure of the nasal septum,to explore its related research on the ability of bone marrow mesenchymal stem cells to form cartilage,and to provide new ideas for the treatment of clinical nasal septum perforation.Methods:1.Extraction,culture and passage of rabbit bone marrow mesenchymal stem cells;observe the cell morphology by HE staining.2.Studies on TGF-?1 on cartilage formation ability of rabbit bone marrow mesenchymal stem cells:CCK-8 was used to determine the effect of TGF-?1 on rabbit bone marrow mesenchymal stem cells proliferation;toluidine blue,safranin O,and alixin blue staining,Type ? collagen immunohistochemical observation of cartilage differentiation ability.3.Preparation of TGF-?1 concentration gradient hydrogel scaffolds for the characterization of materials,biocompatibility of materials,and related studies on the release rate of materials.4.TGF-?1 concentration gradient hydrogel scaffolds observe cell adhesion by fluorescent staining,observe cartilage proteoglycan secretion in materials by toluidine blue and alixin blue staining,observe formation of cartilage matrix by safranine,and observe by immunohistochemistry of collagen type ? and expression of PCR gene level to evaluate chondrocyte production.Results:1.Rabbit bone marrow mesenchymal stem cells are generally spindle-shaped and vortex-shaped;fifth-generation cells have the strongest proliferation capacity and stable differentiation capacity.2.CCK-8 showed that TGF-?1 has the ability to proliferate on rabbit bone marrow mesenchymal stem cells;TGF-?1 secretes proteoglycan,chondrocyte matrix,and type ?collagen to rabbit bone marrow mesenchymal stem cells to promote its differentiation into chondrocytes.3.The TGF-?1 concentration gradient hydrogel scaffold is a porous scaffold,which is biocompatible,does not cause toxic and side effects to the cells,and can stably and continuously release TGF-?1 to form a stable cell differentiation concentration,so that Rabbit bone marrow mesenchymal stem cells were largely differentiated into chondrocytes.Conclusion:TGF-?1 concentration gradient hydrogel scaffold material has stable biodescriptive and porous scaffolding effects,can promote the increase of chondrocytes,and has a strong promotion effect on cartilage repair.It can open up new treatments for clinical nasal septum perforation.
