Objective:To observe the expression and changes of triglyceride and cholesterol content before,during and after modeling after TGF-?1 intervention in rabbit with steroid-induced femoral head necrosis(SANFH);spiral CT and light microscopy were used to detect the pathological changes of femoral head bone tissue in New Zealand white rabbits,and then to observe the efficacy of TGF-?1 in the treatment of rabbit SANFH and explore its mechanism of action.Method:Forty-four New Zealand white.rabbits were randomly distributed into control group(8),model group(18)and treatment group(18),both the model group and the treatment group started modeling at week 2.During the experiment,the control group was fed normally;model group:normal feeding after modeling;treatment group:after modeling,TGF-?1 was intramuscularly injected and normally fed.The serum triglyceride and cholesterol levels of rabbits were measured at the corresponding time points before,during and after modeling,and the bone tissue samples of bilateral femoral head were observed under the microscope,and the cavity rate of each group was calculated.Results:1.Behavioral changes of New Zealand white rabbits:before modeling,New Zealand white rabbits were in good mental condition,had normal diet,glossy hair,and normal limb joint movement.After modeling,the New Zealand white rabbits suffered from inactivity,decreased appetite,delayed reaction,dull hair,decreased muscle strength of hind limbs and decreased activity,and some New Zealand white rabbits showed lameness and other early SANFH symptoms.After treatment with TGF-?1,at the same time point,the muscle strength,range of motion and claudication of the limbs after treatment with TGF-?1 in the treatment group were improved compared with the model group,but still decreased compared with the control group.With the passage of time,the early SANFH symptoms in the treatment group all recovered to varying degrees.2.Triglyceride and cholesterol:there was no significant difference in triglyceride and cholesterol before modeling New Zealand rabbits(P>0.05).At the end of the third week,compared with the control group,both the model group and the treatment group had higher TC,TG and TG levels(P<0.05).At the end of the 7th week,TC and TG in the treatment group were lower than those in the model group,but still higher than those in the control group(P<0.05).3.CT imaging examination:at the end of week 7,the control group:imaging examination showed smooth surface and complete appearance of bilateral femoral head,and no obvious abnormal changes were observed inside the femoral head.At the end of week 7,the model group:imaging examination showed that the surface of the femoral head was not smooth,the femoral head collapsed,the shape was irregular,and there were cystic changes inside the femoral head and low density areas.Treatment group at the end of the 7th week:imaging examination showed that the surface of bilateral femoral head was slightly uneven,no femoral head collapse was observed,and no obvious abnormal signals were found inside the femoral head.4.The empty bone lacuna rate:No abnormal pathological changes were observed in the control group in HE staining.At the end of the third week,both the model group and the treatment group had osteocyte lacunae.Under the same sampling time,compared with the model group,the number of osteoblasts in the treatment group decreased to different degrees(P<0.05).As time went on,the number of empty bone lacunae of osteocytes in the treatment group at the end of the 7th week decreased to different degrees compared with that at the 3rd week(P<0.05).Conclusion:1.This experiment proved that the rabbit SANFH model could be established successfully by lipopolysaccharide combined with large dose of hormone.2.TGF-?1 has a obvious effect on rabbit SANFH,improving the symptoms of rabbit SANFH and alleviated the course of femoral head necrosis in rabbits.3.Triglyceride and cholesterol are closely related to the pathological process of SANFH.Lowering blood triglycerides and cholesterol may be one of the mechanisms by which TGF-?1 interferes with SANFH.Figure[4]table[4]reference[87].
