Objective:1.To investigate the difference of expression of RNA N6-methyladenosine Modification and its catalytic gene METTL14 in laryngeal carcinoma tissue,Mild dysplasia,normal laryngeal mucosa(including inflammatory tissue)and its correlation with the clinicopathological characteristics of laryngeal carcinoma.2.To investigate the recovery of patients with laryngeal carcinoma who were treated with METTL14,one of the components of N6-methyladenosine modified methyltransferase,it provides a theoretical basis for studying the pathogenesis and initial diagnosis of squamous-cell Carcinoma.Methods:1.To collect the medical records of 60 patients with laryngeal carcinoma between 2017 and 2019 in the Otolaryngology Head and Neck Surgery department of the affiliated hospital of Northern Sichuan Medical College;2.To collect 20 cases of polyp and inflammatory tissue,20 cases of mild dysplasia,corresponding wax mass in 60 patients with Laryngeal Carcinorna,and 15 cases of recurrent laryngeal cancer patients in 2019,10 cases of fresh and normal laryngeal tissue of laryngoscope in outpatients in 2019;3.The expression of the catalytic gene MFTTL14 in Laryngeal Carcinoma,atypical tissue and normal laryngeal Mucosa was examined by immunohistochemical SABC two-step method,The expression intensity of METTL14 in the fresh frozen tissues of 15 recurrent laryngeal squamous cell carcinoma(LSCC)was semi-quantitatively detected by real-time polymerase chain reaction(rt-PCR);4.To compare the expression level of METTL14 in laryngeal cancer cells of patients with recurrent laryngeal cancer in 2019 who were diagnosed and treated for the first time from 2017 to 2019 with other laryngeal cancer patients in the same period,and to analyze the relationship between METTL14 expression and recurrence,and statistical analysis of the data,The relationship between the expression of METTL14 and the clinicopathologic features of the patients with primary laryngeal carcinoma was analyzed;5.The statistical method uses SPSS20.0 to analyze the data,and P<0.05 is statistically significant.Results:Immunohistochemical and RT-PCR were used to analyze the collected samplesThe results showed that the expression of METTL14 was high in both primary and recurrent laryngeal squamous cell carcinoma(LSCC),but low or no expression in mild dysplasia and normal laryngeal tissues;however,there was no difference in the expression of METTL14 between primary and recurrent laryngeal squamous cell carcinoma(P>0.05);The expression of METTL14 is closely related to the clinical stage,differentiation and lymph node metastasis of laryngeal carcinoma.Conclusion:The high expression of METTL14 in Laryngeal squamous cell carcinoma(LSCC)suggests that it may be an early diagnostic marker of LSCC.Secondly,METTL14 was positively correlated with the clinical stage of laryngeal Carcinoma and negatively correlated with the degree of differentiation of Laryngeal Carcinoma.It suggests that MFTTL14 is associated with the progression of Laryngeal squamous Cell Carcinoma,and can be used as an index for evaluating the treatment and prognosis of LSCC.Mettl 14 may provide a therapeutic target for Laryngeal squamous Cell Carcinoma.
