Objective:To study the effects of C/EBP? overexpression and interference on the proliferation of human colorectal cancer cells in vitro and the growth of tumor implants in nude mice,tumor volume changes and tumor weight.Methods:1.1.Construct a lentiviral expression vector containing C/EBP-?overexpression and C/EBP-? interference,and transfect human colon cancer Caco-2 cell line.Observe whether the fluorescence is expressed by a fluorescence microscope to determine whether the transfection was successful.2.Human colon cancer Caco-2 cell line transfected with C/EBP-? overexpression or C/EBP-? interference lentiviral vectors,CCK8 kit was used to detect the effects of C/EBP-? overexpression or C/EBP-? interference with lentiviral vectors on cell proliferation.3.A human colon cancer Caco-2 cell line transfected with C/EBP-?overexpression and C/EBP-? interference lentiviral vectors to establish a model of subcutaneous tumor formation in nude mice to detect C/EBP-? in vivo Effects on tumor growth,tumor volume,and tumor weight.Results:1.Compared with the normal control group,there was no significant change in cell proliferation in the overexpression no-load transfection group and the interference no-load transfection group;Compared with the overexpression no-load transfection group,the cell proliferation ability of the C/EBP-? overexpression transfection group was reduced;Compared with the interference-free transfection group,the cell proliferation ability of the C/EBP-? interference-transfected group was increased.2.Compared with the normal control group,there was no significant change in the tumorigenicity of cells in the over-expression no-load transfection group and the interference no-load transfection group;Compared with the overexpressing no-load transfection group,the C/EBP-? overexpressing transfection group had reduced tumorigenicity;Compared with the interference-free transfection group,the C/EBP-?interference-transfected group had an increased tumorigenicity.Conclusion:1.The expression of C/EBP-? has a "negative regulation" effect on the proliferation of human colorectal cancer cells in vitro.2.In nude mice tumor formation models,the expression of C/EBP-? also has a"negative regulation" effect on the tumorigenic capacity of human colorectal cancer cells in vivo.3.The expression of C/EBP-? protein may show a "negative correlation effect" in human colorectal cancer,and through the relevant mechanism of action,it may inhibit the growth of human colorectal cancer;and may be further used as a relevant therapeutic target for the treatment of human colorectal cancer.
