Thrombotic diseases are a kind of diseases that seriously endanger human life and health.It can affect all organs and systems throughout the body,and its morbidity,mortality,and mortality are very high.Studies have shown that the pathogenesis of thrombosis is mainly related to vascular factors,thrombosis,changes in physical and chemical properties of blood and changes in blood flow.The current clinical treatment of thrombotic diseases is mainly aspirin and clopidogrel,but side effects such as drug resistance,severe bleeding,and non-specific selection may occur after long-term use.Polygala tenuifolia Willd.Seeding is derived from the aerial part of Polygala tenuifolia Willd.or Polygala sibirica L.,A plant of Polygala tenuifolia Willd.,and has antioxidant,anti-inflammatory and other activities.In addition,the plant of the same genus melon seed has the effect of promoting blood circulation and removing blood stasis,and it is mixed with the whole plant of Polygala tenuifolia Willd.in North China,so it is speculated that Polygala tenuifolia Willd.Seeding.may have anti-platelet aggregation activity.Therefore,this study uses Polygala tenuifolia Willd.Seeding as the material,using HPLC and UHPLC-TSQ-MS to determine and identify the content of Polygala tenuifolia Willd.Seeding,crude extract of Polygala tenuifolia Willd.Seeding.,and polyphenols of Polygala tenuifolia Willd.Seeding.The anti platelet aggregation and anti thrombosis effects of Polygala tenuifolia Willd.Seeding were determined by anti platelet aggregation experiment in vitro and thrombus model in vivo,and the mechanism of action was studied preliminarily.The main content and results include:1. Using ADP as an inducer,the in vitro platelet aggregation inhibitory activity was determined on the crude extracts of the Polygala tenuifolia Willd.Seeding,polyphenols of the Polygala tenuifolia Willd.Seeding,mangiferin and 3,6'-dibrevisyl sucrose.,and it was found that polyphenols of the Polygala tenuifolia Willd.Seeding had better resistance to ADP as an inducer than crude extracts of the Polygala tenuifolia Willd.Seeding,mangiferin and 3,6'-dibrevisyl sucrose.Comparing the inhibitory effects of polyphenols of the Polygala tenuifolia Willd.Seeding on adenosine diphosphate?ADP?,arachidonic acid?AA?,collagen?COLL?,and epinephrine?EPI?inducers,it was found that the effect of Polygala tenuifolia polyphenols on ADP was better than that of AA,Coll and EPI,suggesting that the main pathway of the inhibition of platelet aggregation was ADP and AA.2. In vivo pharmacodynamic experiments,observations were made of the Polygala tenuifolia Willd.Seeding polyphenols?coagulation?through three models of mouse tail vein blood clotting time,collagen-epinephrine-induced acute pulmonary thrombosis in mice,and common carotid artery thrombosis in rats)The effects of blood time,hemiplegia and death in mice,and thrombus length and mass in rats were evaluated on the antithrombotic activity of the Polygala tenuifolia Willd.Seeding polyphenols.The results show that the Polygala tenuifolia Willd.Seeding polyphenols can prolong the tail vein bleeding time in mice;Reducing wet weight and length of thrombus in common carotid artery of rats,significantly increase the production of 6-keto-PGF1aand t-PA,and reduce TXB2and PAI-1 level.It is suggested that polyphenols from Polygala tenuifolia Willd.Seeding can inhibit thrombosis by increasing the production of 6-Keto-PGF1aand t-PA,and decreasing the levels of TXB2 and PAI-1.3. A model of acute blood stasis in rats was induced by subcutaneous injection of high-dose epinephrine hydrochloride plus ice water bath.The drug was administered orally by gavage.for 7 days,The blood was taken from the common carotid artery 0.5 h after the last replacement,and the whole blood viscosity,plasma viscosity and red blood cell results were measured.The results show that Polygala tenuifolia Willd.Seeding polyphenols can improve the abnormal blood rheology,at the same time widen the myocardial cells of acute blood stasis rats,and pulmonary interstitial edema has improved to a certain extent.This may be related to Polygala tenuifolia Willd.Seeding polyphenols can significantly reduce the content of TXB2and ET-1 in the blood and increase the content of 6-keto-PGF1aand e NOS in the blood.
