Objective: To confirm the effect of oscillating electric field stimulation on regulating the directional differentiation of transplanted neural stem cells and promoting the recovery of nerve function after spinal cord injury,to provide theoretical reference for the study of its mechanism,and to explore the new technical direction of repairing spinal cord injury.Methods: SD rats were randomly divided into SCI group,SCI+NSC group and SCI+NSC+OFS group,and the SCI rat model was constructed by using NYU hammer.BBB score was used to evaluate neurological function recovery after SCI.Frozen sections,Western Blot and Immunofluorescence were used to observe the proliferation and differentiation of neural stem cells transplanted into the injured spinal cord of rats under the action of OFS.The damage and regeneration of axons after spinal cord injury in rats were detected by WB and IF.Results: 1.The results of BBB score showed that the mean score of the rats in SCI+NSC+OFS group was significantly higher than SCI+NSC group at all time points 3 to 5 weeks after surgery(P<0.05);2.WB results showed that the expression of GFP in the spinal cord of rats in SCI+NSC+OFS group was significantly higher than SCI+NSC group on the 3rd day after surgery(P<0.05);3.IF results showed that the number of GFP/?-tubulin III positive cells in the spinal cord of SCI+NSC+OFS group was significantly higher than SCI+NSC group at the 3rd,7th and 14 th day after surgery(P<0.05);4.IF results showed that the number of GFP/ NG2 positive cells in the spinal cord of SCI+NSC+OFS group was significantly higher than SCI+NSC group on the 3rd,7th and 14 th day after surgery(P<0.05);5.IF results showed that the number of GFP/ S100?positive cells in the spinal cord of SCI+NSC+OFS group was significantly lower than SCI+NSC group on the 7th and 14 th day after surgery(P<0.05);6.WB results showed that the expression of NFH in the spinal cord of rats in SCI+NSC+OFS group was significantly higher than that in SCI+NSC group on the 14 rd day after surgery(P<0.05),IF results showed that the number of NF-H positive area in the spinal cord of SCI+NSC+OFS group was significantly bigger than SCI+NSC group on the 14 th day after surgery(P<0.05).Conclusion: OFS can regulate the directional differentiation of NSC transplanted in rats,promote the regeneration of neurons,the re-myelination of nerves and the regeneration of axons,reduce the formation of astrocyte scar;Compared with NSC transplantation alone,combination therapy was more beneficial to the recovery of nerve function in rats after SCI.
