Objective:Oral squamous cell carcinoma(OSCC)is one of the most common malignancies of the head and neck,accounting for more than 90% of all oral cancers and the sixth most common type of cancer in the world.The disease has a variety of factors,including high incidence,strong local invasiveness and high rate of cervical lymph node metastasis.In addition to surgical treatment,adjuvant chemoradiotherapy can be selected according to its staging.The occurrence and development of oral cancer is complex,which is regulated by gene level,gene expression is regulated by DNA level,transcriptional level,post-transcriptional level and protein level.The analysis and use of valuable markers mainly involved in the pathogenesis of oral squamous cell carcinoma can help us to understand the disease in a more detailed and comprehensive way,and then we can choose to implement targeted,personalized,practical and effective treatment.Therefore,in order to improve the survival rate and quality of life of oral squamous cell carcinoma patients,we need to find effective tumor affecting factors,new treatment methods and anti-tumor drugs.As an important part of innate immunity,antimicrobial peptides(AMPs)can play a variety of roles,such as anti-infection,anti-endotoxin,chemotaxis,immune regulation and anti-tumor effects.LL-37 is the only antibacterial peptide in the human body,studies have shown that it for a variety of cancer cells,for ovarian cancer,lung cancer,breast cancer,prostatecancer,pancreatic cancer,malignant melanoma(MM)and skin squamous cell carcinoma(SCC)may promote the growth and metastasis of cancer cells,but in malignant tumor,colon cancer,gastric cancer,blood system in oral squamous cell carcinomas,such as tumors have certain anti-cancer effect.The research on the effect of LL-37 on oral squamous cell carcinoma may provide new ideas and methods for the treatment of oral squamous cell carcinoma.Oral squamous cell carcinoma cells cultured in vitro were treated with different concentrations of antimicrobial peptide ll-37,The results were analyzed to investigate the effects of different concentrations of antibacterial peptide ll-37 on the proliferation,invasion and apoptosis of oral squamous cell carcinoma cells,in order to further evaluate the application of ll-37 in the prevention and treatment of oral infection and tumor,a certain basis for laboratory research was provided.Method:OSCC cells were cultured in vitro and treated with different concentrations of antimicrobial peptide LL-37.A blank control group and groups with different concentrations of antimicrobial peptide LL-37 were set.Cell proliferation was detected by CCK8,apoptosis was determined by Annexin V/PI double staining,and the effect of LL-37 on OSCC cell invasion and migration was detected by transwell assay.Result:1.Cell proliferation-toxicity assay(CCK8 assay): after the treatment of cells with different concentrations of LL-37,the cell proliferation was weakened,and the inhibitory effect was positively correlated with the concentration of LL-37 to some extent.2.Apoptosis experiment(Annexin V/PI staining assay): The apoptosis rate of cells increased after treatment with LL-37 concentrationin each group.3.Cell invasion experiment(transwell experiment): compared with the control group,after the cells were treated with different concentrations of LL-37,the number of cells invading into the lower chamber in the transwell chamber decreased,indicating that the cell invasion ability was weakened by the action of LL-37.4.ELISA: compared with the control group,the amount of TNF-?,IL-6 and IL-8 secreted by cells treated with the increased of LL-37.Conclusion:Antimicrobial peptide LL-37 inhibits the proliferation and invasion of OSCC cells and promotes the apoptosis of cells.
