| Objective:Based on proteomics,we studied the expression of mitochondrial differentially expressed proteins in liver of mice with natural growth of 16 months old,explored the protein markers and pathways related to aging,and regulated the protein markers and pathways of Liuwei Dihuang pills.To provide basis for elucidating the mechanism of aging process and the biological basis of Liuwei Dihuang Pill intervention and natural aging-related syndromes in miceMethod:This experiment selected 60 mice of grade SPF ICR as the experimental research obj ect,the natural aging 16 months old mice were randomly divided into model group and administration group,each group of mice 10 males,10 females,another 3 month old male mice 10,female mice 10 as blank group,blank group and model group with the same volume percentage of normal saline gavage,the administration group with Liuwei Dihuang pills.The mitochondrial protein markers of liver tissue and the related protein markers of Liuwei Dihuang pills in mice with natural aging model were selected by UniProt GO analysis and KEGG analysis techniqueResult:1.The model group compared the protein markers of hepatic mitochondria sample in the blank group and screened out nuclear pore complex protein Nup98、myosin light chain 3,cytochrome P450.4A10 and other related differential protein markers and insulin signaling pathway,endophagy,metabolic pathway and other biological pathways2.Liuwei Dihuang Pill regulates differential proteins during natural aging,including ribosomal binding protein 1,nuclear pore complex protein,NAD kinase,serine/arginine repeat matrix protein 1,integrin α-9,etc3.GO analysis shows that Liuwei Dihuang pills mainly participate in the process of drug metabolism,purine nucleoside monophosphate metabolism,organic matter transport,metabolic process and other biological metabolic processes;participate in the synthesis of mitochondria,endoplasmic reticulum membrane and other important cell components;participate in the complex binding containing a variety of proteins,catalytic activity,redox enzyme activity,actin filament activity binding to regulate molecular physiological functions4.By using KEGG methods to analyze the experimental results,it can be concluded that the main pharmacological effects of liuwei dihuang pills include the physiological function regulation of various related signaling pathways,such as oxidative phosphorylation,insulin signaling pathway,glycine,serine and threonine metabolism,endoplasmic reticulum protein processing,hypertrophic cardiomyopathy and so onConclusion:1.In this experiment,we used proteomics technique to explore the mitochondrial protein markers in liver tissue of natural aging model mice,and to explore and identify the differential protein markers related to aging in Liuwei Dihuang pills2.Compared with the age of 16 months and 3 months,the intervention of Liuwei Dihuang pills can effectively regulate the differential proteins such as ribosomal binding protein 1,nuclear pore complex protein Nup98-Nup96、serine/arginine repeat matrix protein 1,which is mainly enriched in oxidative phosphorylation,insulin signaling pathway,glycine,serine and threonine metabolism,endoplasmic reticulum protein processing,hypertrophic cardiomyopathy and other five signaling pathways closely related to aging,which indicates that the intervention of Liuwei Dihuang pills can effectively improve the various biological changes that may occur due to natural aging and closely related to the yin deficiency in human body. |
