| Purpose: to use 4 t1 breast cancer in mice transplanted tumor building experimental observation Xia Lianhua soup to transplant type of breast cancer in mice tumor growth inhibition,using the in vitro cultivation of human liver cancer cells SMMC7721,breast cancer cells MCF-7 detection Xia Lianhua soup on tumor cell apoptosis related BCL2,caspase 3 protein expression and the effect of preliminary discussion Xia Lianhua soup effects on tumor cell apoptosis,and explore the compatibility of traditional Chinese medicine(TCM)the suitable compatibility and the interaction relationship of antitumor,provide reference for clinical rational drug use.Methods:In vivo experiment: The experiment was randomly divided into model control group,positive control group,normal control group,xia Lianhua decoction high,medium and low dose group.After 20 days of administration,mice in each group were sacrificed the next day,blood was taken from the eyeball for use,and mice in each group were sacrificed by neck fracture.The weight of the mice,tumor weight,thymus,liver and spleen weights were weighed,and tumor inhibition rate,thymus index,liver index and spleen index of mice in different experimental groups were calculated.In vitro experiment: LIVER cancer SMMC7721 cells were cultured in vitro,and morphological changes of human liver cancer SMMC7721 cells were observed by microscope.MTT colorimetry and DAPI staining were used to observe the effect of Xia Lotus Decoction on the apoptosis of liver cancer SMMC7721 cells.Immunofluorescence experiment: Human breast cancer McF-7 cells were cultured in vitro,and the effects of immunofluorescence technology on the expression of BCL2 and caspase-3proteins in tumor cells were observed 24 hours after cell intervention in xia Lianhua decoction group.Results:1.The high dose,medium dose and low dose groups all had significant tumor inhibition effect on mice with breast tumor transplantation,and the tumor inhibition rates were respectively 46.04%,40.82% and 33.45% in the high dose group,medium dose group and low dose group.2.All groups of TCM had different degrees of inhibition of tumor metastasis,of which the xia Lianhua decoction group was the highest,followed by the Xia Lianhua decoction group,and the Xia Lianhua decoction group was less obvious.3.Xia Lianhua decoction can improve the liver/body ratio,spleen index and thymus index of mice,and protect important organs.4.All the drug groups with different concentrations of Xia Lianhua Decoction had inhibitory effect on SMMC7721 cells of liver cancer,showing apoptotic cell morphology under inverted microscope.5.Immunofluorescence assay showed that xiahlianhua decoction could inhibit the expression of BCL2 protein and improve the expression of caspase-3 protein in human breast cancer McF-7 cells.Conclusion:Xia Lianhua Decoction has an obvious antitumor effect on breast cancer transplanted mice,and the mechanism of action may be to inhibit the expression of BCL2 protein and increase the expression of caspase-3 protein. |
