Objective: To detect the expression levels of sialidase NEU1 and transferase-related ST3GAL3,ST6GAL2,ST8SIA2 mRNA between the autism spectrum disorder group(ASD)and healthy control groups;the ASD group heart and spleen deficiency syndrome and heart and liver fire syndrome.To explore the clinical significance of these genes with ASD is important guiding significance for the disease in traditional Chinese medicine(TCM)syndrome differentiation and diagnosis.Methods:(1)Collect peripheral blood of the subjects from May 2019 to December 2019 at the Children's Health Department of Xi'an Children's Hospital.There were 59 ASD children and age-matched healthy children.The ASD group was divided into 32 syndrome of heart-liver fire and 27 heart and spleen deficiency syndrome according to TCM syndrome differentiation.(2)The relative expression of sialidase and transferase-related genes in each group was detected by RT-qPCR.The relative expression of sialidase and sialyltransferase related genes in each group was detected by RT-qPCR.Results:(1)The ASD children peripheral blood relative expression of sialidase NEU1 mRNA [4.559(1.557~6.866)] significantly higher than the healthy control group [1.174(0.740~1.638)],(P<0.05).(2)The ASD children peripheral blood relative expression of sialyltransferase ST3GAL3 mRNA [0.073(0.043~0.169)] significantly lower than the healthy control group [0.231(0.069~0.316)],(P<0.05).(3)The ASD children peripheral blood relative expression of sialyltransferase ST6GAL2 mRNA [0.036(0.012~0.092)]significantly higher than the healthy control group [0.021(0.008~0.062)],(P<0.05).(4)The ASD children peripheral blood relative expression of sialyltransferase ST8SIA2 mRNA [0.082(0.036~0.276)] higher than the healthy control group [0.078(0.040~0.126)],but the difference was not statistically significant(P> 0.05).(5)In the ASD group,the heart and spleen deficiency syndrome peripheral blood relative expression of sialidase NEU1 mRNA [6.343(2.731~9.639)] significantly higher than the heart and liver fire syndrome[2.265(1.322~5.423)](P <0.05).The sialyltransferase ST3GAL3 and ST6GAL2 mRNA were not statistically significant in ASD group between two TCM syndromes.Conclusion: In this experiment,RT-qPCR was used to detect the peripheral blood mRNA expression of sialidase NEU1 and sialyltransferases ST3GAL3,ST6GAL2,and ST8SIA2 in children with ASD and healthy controls.The statistical results show that the technology has certain feasibility in the study of ASD and its TCM syndrome types,and provides a new research idea and method for the occurrence mechanism of ASD and TCM syndrome differentiation.The expression of NEU1,ST3GAL3 and ST6GAL2 mRNA is different between ASD group and healthy control group,suggesting that these genes may become potential objective biomarkers for clinical diagnosis of ASD.It has important guiding significance for clinical screening,diagnosis and treatment of the disease.At the same time,the expression of mRNA of NEU1 different between the two syndromes of heart and liver fire syndrome and heart and spleen deficiency syndrome in the ASD group.It may be related to the microscopic material expression of different TCM syndromes based on molecular biological basis,which will help to become a potential specific indicator to distinguish the two TCM syndromes. |
