Obesity is defined as abnormal or excessive fat accumulation.It is a major inducer of many chronic diseases.Black soybean has many effects as a homologous and an important raw material for medicine and food.Existing researches mainly focus on the research on the health functions and pharmacological activities of black soybean,while the research on black soybean coat is relatively rare.It is of great significance for improving obesity and increasing the utilization value of Qingren black soybean coat to study the effect of Qingren black soybean coat extract on weight inhibition in obese mice.In this paper,Qingren black soybean(Glycine soja Sieb.Zucc)coat was used as the research object.Ethanol extract and water extract were obtained by different reagent extraction methods,and the differences in the components of the two extracts were analysed by using Liquid Chromatography-Mass Spectrometer(LC-MS).In order to study the inhibitory effect of extract on the weight of diet-induced-obeity mice,the model of C57BL/6J mice was established by feeding high-fat diet,and used black soybean coat extract for treatment to study the inhibitory effect of extract on the weight of obese mice.After that,blood samples and liver samples were collected from the mice.The serum triglyceride levels in mice were detect by the kit.Liver sections of mice were prepared for hematoxylin-eosin staining to analyze the effects of the extract on the liver fat of obese mice.The toxicity of the extract was tested by vitro cell experiment and acute toxicity test in mice.LC-MS result showed that there were some differences in the composition of the two extracts.The ethanol extract contained resveratrol,cyanidin-3-O-glucoside,peonidin-3-O-glucoside,pelargonidin-3-O-rutinoside,catechin,delphinidin-3-O-glucoside and phloretin 2'-O-?-L glucopyranoside.And the water extract contained resveratrol,but there was no other components mentioned above.The experimental results of the inhibitory effect of the extract on weight showed that the extract had an inhibitory effect on the weight of obese mice with normal diet or high-fat diet.In the experimental groups on a high-fat diet,the average weight of mice in the control group increased 5.05 g average,the average weight of mice treated with ethanol extract decreased 1.24 g,and the average weight of mice treated with water extract decreased 0.56 g.Histological results of the liver showed that compared with the control group,both the black soybean extract and water extract reduced the liver fat of the mice.Compared with serum triglyceride levels(2.47 mmol/L)in the control group,the black soybean coat extract reduced the triglyceride levels of the mice serum.The triglycerides of the ethanol extract intervention group and the water extract intervention group were 1.35 mmol/L and 1.44 mmol/L,respectively.The result of MTT showed that different concentrations of extract were applied to HL-7702 cells for 48 h,the cell survival rate increased with the increase of the concentration.In the acute toxicity testin vivo,the dose of the ethanol extract was 5000 mg/kg,there had no mouse died.The results of extract toxicity test showed that black soybean coat extracts were safe and non-toxic.These results in this paper were expected to increase the added value of Qingren black soybean coat.It will provide a plant extract which is potentially effective,new,natural,safe.It could improve obesity and provide a basis for subsequent research.
