Cancer poses a huge threat to human health.Cytological diagnosis is an important cancer diagnosis technology.The HE(Hematoxylin-Eosin)staining effect of the tissues or cells pathological section directly affects the decision made by the pathologist.The factors that have the greatest impact on the staining effect are the dye solution concentration and dyeing temperature.To develop dye concentration detection and temperature control module is of great practical significance and value for the best dyeing effect.This paper designed an automated and low-cost concentration detection module to detect the dye concentration.The absorbance method based on Lambert 's law.Use optical path to detect changes in dye concentration,including LED light source emitting fixed wavelength,light source driving circuit based on CAT4010,low-light detection system based on photoelectric sensor S1226-5BQ and logarithmic amplifier LOG112,and analog-to-digital converter based on AD7705.Besides,different dyes require different temperature,a temperature detection and control module is developed to ensure dye activity,circuit which based on DS18B20 and Peltier is designed.Fuzzy adaptive PID control algorithm is adopted to overcome the time lag and inertial characteristics of temperature.The concentration and dyeing time,concentration and optimal dyeing temperature data obtained by many experiments are fitted with a least squares curve fitting method to establish a model of concentration and dyeing time at a certain temperature to determine specific staining time according to different concentrations.For dyes of different concentrations,under the condition that the dyeing time remains unchanged,the dyeing temperature can be increased to compare the dyeing effect of slices at different temperatures,and the optimal dyeing temperature can be selected to obtain the function curve of the concentration and temperature.The appropriate dyeing temperature can be set according to the concentration of the dye.The experiment shows that all functions of the concentration detection and temperature control modules have been successfully realized and put into use.They are a major technological breakthrough in improving the performance of automatic dyeing machines.
