In cancer cells,changes in glycosylation on proteins are often detected.When tumors occur,abnormal glycosylation results in changes in the structure or number of sugar chains on glycoproteins,as well as on lipid molecules.This phenomenon leads to changes in the sugar chains in the glycolipid;in addition,the expression of sugar-binding proteins(GBPs)that interact with the sugar chains will also change.The glycoproteins on the cell surface are involved in many important biological processes,and the glycosylation on them has an important effect on the function of glycoproteins;studies have shown that abnormal glycosylation on glycoproteins can cause the invasion and migration of tumor cells.Change the malignant phenotype of other tumors.N-cadherin is a marker of epithelial-mesenchymal transition(EMT)and one of the important members of the cadherin family.Clinical studies have shown that N-cadherin is mainly located in the cell membrane and cell junction Its function is mainly to mediate the adhesion between cells.Although N-cadherin is a typical glycoprotein,however,the research on glycosylation on it is not clear,so it is of great significance to study the specific effect of glycosylation on N-cadherin for cancer.This study analyzed the expression of N-cadherin in the same organs of normal humans and cancer patients based on data,and found that the expression of N-cadherin in some organs of cancer patients changed compared with normal humans.A comparative analysis of the mRNA levels of N-cadherin in hepatocellular carcinoma and adjacent tissues revealed that N-cadherin exhibited high mRNA levels in liver cancer compared with adjacent tissues,but less research on N-cadherin in liver cancer,It is still unclear whether the glycosylation on it has an effect on the canceration process of liver cancer.Subsequently,this study conducted a prognostic analysis of N-cadherin on liver cancer,and explored whether the expression of N-cadherin would affect the prognosis of liver cancer patients.It was found that among liver cancer patients,the survival time of patients with high expression of N-cadherin was higher than that of patients with low expression of N-cadherin.short.In order to better understand the function of N-cadherin in liver cancer,based on the data,an enrichment analysis of N-cadherin-related genes in liver cancer was carried out.First,an analysis of mRNA-level related genes was found,and it was found to exist with N-cadherin in liver cancer There are more than 700 genes related to the mRNA level,and then enrichment analysis of related genes at the protein level,whether at the mRNA level or protein level,enrichment analysis of related genes,N-cadherin-related genes participate in liver cancer In many important biological processes,N-cadherin-related genes are closely related to important pathways such as adhesion pathways and proteasome degradation.To investigate how N-cadherin glycosylation affects liver cancer,this study first analyzed the mutation of N-cadherin in liver cancer and found that the probability of N-cadherin gene mutation in liver cancer is low and the protein structure There are fewer possible mutation sites in the domain,and it is speculated that N-cadherin is more likely to rely on the post-transcriptional modification process in liver cancer.Next,N-cadherin N-glycosylation sites were predicted and found that there were 190,273,325,402,572,622,651,692 eight potential N-glycosylation sites on N-cadherin,Design site-directed mutation primers according to the potential sites predicted by the website,and construct 8 mono-N-glycosylation site mutant recombinants and 8 N-glycosylation site full-mutation recombinants with the pCMV-Flag vector as the empty load.The constructed recombinant plasmid was identified.In summary,compared with normal humans,the expression of N-cadherin in the same organ of cancer patients is different;compared with adjacent tissues,N-cadherin is highly expressed in liver cancer tissues;and N-cadheirn high expression will have an important impact on the prognosis of liver cancer patients;N-cadherin-related genes in liver cancer are closely related to adhesion pathways and proteasome degradation pathways.Through the analysis of N-cadherin gene mutations and protein domain mutations in liver cancer,it was found that N-cadherin may rely on post-translational modifications in liver cancer,so the potential N-cadherin N-glycosylation site mutation recombinant was constructed,they will lay a foundation for the follow-up study of the effect of N-glycosylation on N-cadherin on liver cancer cells.
