Objective To investigate the expressions of E-cadherin and N-cadherin in benign prostatic hyperplasia tissues and prostate cancer tissues.To detect the function and significance of E-cadherin and N-cadherin in human prostate cancer progression.Method The expression of E-cadherin and N-cadherin in51tumor tissues and47prostatic hyperplasia tissues were determined by immunohistochemical method. To determine positive results of E-cadherin and N-cadherin using semi-quantitative integration method. According to tumor differentiation grade and clinical stage,we analyzed the relationship among E-cadherin, N-cadherin and prostate cancer by using x2test.RT-PCR was used to detect the expression of E-cadherin mRNA and N-cadherin mRNA in benign prostatic hyperplasia and prostate cancer and analyzed statistically.Result1.In benign prostatic hyperplasia tissues and prostate cancer tissues,the positive expression rate of E-cadherin are87.23%and31.37%,and the positive expression rate of N-cadherin are4.26%and86.27%.The expression of E-cadherin and N-cadherin in prostate cancer tissues are statistically significant comparing with their expression in benign prostatic hyperplasia tissues(P<0.01).2. The positive expression rates of E-cadherin were75%,46.67%and10.71%in high-differentiation group, middle-differentiation group and low-differentiation group in prostate cancer tissues; The positive expression rates of N-cadherin were37.5%,80%and96.43%in high-differentiation group, middle-differentiation group and low-differentiation group in prostate cancer tissues; The expression of E-cadherin and N-cadherin between all groups are statistically significant(P<0.05).3.The positive expression rates of E-cadherin were100%,75%,36.81%and4.55%in every clinical stage of prostate cancen:A、B、C and D stage. The positive expression rates of N-cadherin were50%,37.50%,57.89%and86.36%in every clinical stage of prostate cancerA、B、C and D stage.4.The expression of E-cadherin mRNA and N-cadherin mRNA in prostate cancer with low-differentiation and benign prostatic hyperplasia,prostate cancer with middle-differentiation and benign prostatic hyperplasia were statistically significant(P<0.05),while The expression of E-cadherin mRNA and N-cadherin mRNA in prostate cancer with high-differentiation and benign prostatic hyperplasia was not significant. Conclusion1. The expression of E-cadherin is negatively associated with the malignancy and clinical stage of prostate cancer.2. The expression of N-cadherin is positively associated with the malignancy and clinical stage of prostate cancer.
