The process of ejaculation is mainly regulated by the brain and spinal cord.Monoamine neurotransmitters,especially 5-HT,take a significant role in the regulation of ejaculation.5-HT functions by connecting to different receptors on the post-synaptic membrane.SERT,located in the presynaptic membrane,can re-uptake 5-HT in the prominent space and participate in the regulation of ejaculation.It is an important target for studying ejaculation regulation.Purpose:This study is to explore changes in sexual behavior and other neurotransmitters and5-HT receptor that may occur after SERT knockout in rats.And we are further aimed to understand the relationgship between SERT and ejaculation regulation.Materials and Method:By target-selected mutagenesis,we generated the SERT-/-,SERT+/-and SERT+/+ male rats,which were divided into three groups.There are 4 rats in each group.Male rats of the three groups and female rats after estrus treatment were subjected to mating experiments in the same cage.At the same time the sexual behavioral indexes of male rats were recorded.After mating experiments,male rats were sacrificed and the brain and spinal cord tissues to be measured were cut out and preserved well.The tissues were dissected and used for SERT,5-HTR1 A,5-HTR2 c,TPH2,SERTm RNA,5-HTR1 Am RNA,5-HTR2 Cm RNA,TPH2 m RNA,5-HT,DA,NE examination using QRT-PCR,immunohistochemistry,Western Blot and ELISA techniques.Result:According to the analysis of sexual behavior,SERT-/-male rats completed a longer ejaculation latency and fewer ejaculations compared with SERT + / + male rats.AndSERT +/-male rats also have differences in ejaculation behavior,but the difference are not obvious.As for biochemical characterization,the expression of SERT m RNA,5-HT2 CRm RNA,TPH2 m RNA and SERT,TPH2,5-HT2 CR protein,and DA in related tissues of SERT-/-male rats was significantly reduced compared with SERT + / + in male rats.And the expression of 5-HT1 AR protein,5-HT,NA was significantly increased in SERT-/-male rats.SERT +/-male rats did not change significantly.Conclusion:Our experiments reveal that 5-HT is an important substance involved in the regulation of ejaculation,and SERT is a key substance for 5-HT reuptake.The binding of 5-HT to its receptor mediates the regulation of ejaculation.And when SERT is knocked out,its receptors also change accordingly.There is a certain relationship between DA,NA and 5-HT,which jointly regulate ejaculation.In addition,the differential distribution of SERT and 5HT receptor(5-HTR1 A,5-HTR2C)in brain and spinal cord tissue deserves our further exploration.
