Objective:To analyze the correlation between the expression change of miR-373 in cholangiocarcinoma tissues and clinical stage,and observe the effect of miR-373 on the proliferation and apoptosis of cholangiocarcinoma cells,so as to provide new evidence for the diagnosis and treatment target of miR-373 for cholangiocarcinoma.Methods:1.Collect bile duct carcinoma in patients with tumor and the tumor tissue adjacent to carcinoma,a total of 32,using real-time fluorescent quantitative PCR(qPCR)this was detected in 32 of specimens in the expression of miR-373,and corresponding specimens collected clinical data,including age,sex,tumor TNM staging and presence of lymph node metastasis),and statistical analysis of miR-373 in bile duct carcinoma tissue and corresponding beside the expression differences in tumor tissue and the correlation with bile duct carcinoma TNM staging.2.In order to verify the expression and function of miR-373 in cholangiocarcinoma,normal bile duct epithelial cells(HIBEpiC)and 4 common human bile duct cancer cell lines(RBE,QBC939,HCCC9810,and HUCCT-1)were selected and the expression of miR-373 in human bile duct cancer cell lines and normal human bile duct cancer cell lines were detected by q PCR.3.A cell line with the lowest expression of miR-373 was selected from the above cholangiocarcinoma cell lines as a follow-up functional test cell line.The experimental groups were Control,NC mimic,miR-373 mimic,NC inhibitor,and miR-373 inhibitor.Results:1.Fresh samples of 32 patients with cholangiocarcinoma and adjacent tissues were collected,and the results of real-time quantitative PCR(qPCR)showed that the expression level of miR-373 in the tumor tissues was significantly lower than that in the adjacent tissues.There was no difference in age and gender among 32 patients,and the expression level of mi R-373 was not related to the lymph node metastasis of cholangiocarcinoma,while the expression level of miR-373 in cholangiocarcinoma I/II was significantly higher than that in cholangiocarcinoma III/IV.2.Normal bile duct epithelial cells(HIBEpiC)and 4 common human bile duct cancer cell lines(RBE,QBC939,HCCC9810,and HUCCT-1)were selected and the expression of miR-373 was detected by real-time fluorescent quantitative PCR.When miR-373 was overexpressed,it could significantly inhibit cell proliferation and accelerate apoptosis of the cholangiocarcinoma cell line(HCCC9810).Conclusion:1.miR-373 is poorly expressed in bile duct cancer cells,and the expression level is different in the TNM staging of bile duct cancer.mi R-373 can be used as a index for preoperative diagnosis and evaluation of bile duct cancer.2.The overexpression of mi R-373 can inhibit the proliferation of bile duct cancer cells and accelerate the apoptosis of bile duct cancer cells,which can provide a new idea for the targeted treatment of bile duct cancer.
