Objective : To investigate the effective of celecoxib on proliferation and apoptosis in human cholangiocarcinoma cells line QBC939 in vitro.Methods:1.QBC-939 cells were cultured routinely;2.Cultured cells were divided into 4 groupsxontrol group, 5-Fu group , celecoxib group, celecoxib and 5-Fu drug alliance group; After transfection for 24h,48h,72h 96h,cultured cells were harvested to carry on the next tests;3.To detect the inhibition of human cholangiocarcinoma cells which are treated with celecoxib and 5-Fu by MTT;To detect apoptosis index and cell cycle arrest by flowcytometer.The expression of P21 protein was determined with cell immunohistochemistryResults:1.Different concentration of celecoxib and 5-Fu inhibit proliferation of cholangiocarcinoma cells line QBC939.The inhibitation depends on time and concentration(P<0.01).2.The apoptosis index of QBC939 gradually increase with the increase of drug concentration(P<0.01), S stage cell decreases gradually while G1 stage cell increases(P<0.05),G2 stage cell ratio has no obvious change(P>0.05).3. The expression of P21 protein in human cholangiocarcinoma cells line which are treated with celecoxib and 5-Fu increase(P<0.05)Conclusions :1. celecoxib and 5-Fu inhibits proliferation of cholangiocarcinoma cells line QBC939 in vitro.celecoxib and 5-Fu can induce cholangiocarcinoma cells line QBC939 apoptosis in vitro. celecoxib and 5-Fu drug alliance is more effective than celecoxib or 5-Fu single drug.2. celecoxib and 5-Fu arrest cell cycle of QBC939 cell line in G1 stage. celecoxib and 5-Fu inhibts the growth of QBC939 cells by means of G1 phrase cell-cycle arrest resulting from up-regulation of p21 protein...
