| The neurotoxicity of acrylamide mainly causes slow down of nerve conduction and damage of learning and memory ability.The mechanism of the neurotoxicity is related to the influence of synaptic neuroplasticity,the promotion of hippocampal neuron apoptosis,and the increase of oxidative stress.In China,the content of acrylamide,rice,flour and dried fruit in potato,corn and convenient food is relatively low.Children and adolescents are high-risk groups exposed to acrylamide,and the effect of long-term low-dose acrylamide exposure on learning and memory and higher cognitive function of children and adolescents is not yet clear,which is worth exploring and clarifying the mechanism,and providing experimental reference for the identification and control of food safety risks for children and adolescents.This research adopts the weanling rats as the research object,by gastric doses of acrylamide,compared with the control group by gastric acrylamide 4 to 8 weeks after significantly reduced the weight gain in rats(p < 0.05),the effect is obvious dose-response relationship,acrylamide could seriously affect the growth and development of the juvenile rats should focus on children and adolescents,strengthen health education,lower levels of acrylamide intake.Compared with the control group,after the administration of acrylamide for 8 weeks in weaned rats,blood routine examination showed significant increase in white blood cells,lymphocytes,neutral cells and monocytes(p<0.05),indicating that significant inflammatory reactions may have occurred in rats.Acrylamide exposure increased platelet count(p<0.05),while platelet activation may increase blood viscosity,increase the incidence of cardiovascular disease and promote the development of cardiovascular disease,which may be related to the increased incidence of cardiovascular disease with acrylamide.By analyzing water maze Morris water maze experiment,compared with control group,weanling rats lavage 100 mg/kg acrylamide in rats after 8 weeks after the platform number,platform movement distance,residence time,platform area target area residence time,distance of the target area were significantly lower,show that pubertal rats acrylamide exposure after senior significantly impaired cognitive function of learning and memory.KEGG concentration showed that phenylalanine metabolic pathways,dopaminergic synapses,serotonin can synapses,GABA synaptic,than can synaptic glutamate neurotransmitter pathways and represented by PI3K-Akt signaling pathways of inflammation reaction pathways were obvious influence,dopaminergic neurotransmitter dopamine synaptic material L-tyrosine content decreased obviously,the precondition of serotonin can synapses a precursor of the neurotransmitter serotonin levels of tryptophan decreased obviously.Acrylamide interference neurotransmitter precursors of synthetic metabolism,neurotransmitter receptors interference Gi/o protein receptor expression that cause imbalance of LTP and LTD,synaptic plasticity is severely affected,phenylalanine metabolism,dopaminergic synapses and synaptic abnormal metabolic pathways may be serotonin can acrylamide influence higher cognitive functions of the important mechanisms of learning and memory. |
