Objective:This study was based on Genomic Wide Association Studies of European populations with blood pressure traits to investigate whether three new single nucleotide polymorphisms(VEGFA rs9472135,FGD5 rs11128722,and ZC3HC1 rs11556924)associated with endothelial dysfunction might also have an impact on the incidence of essential hypertension in elderly populations in Liaoning province.This will provide new therapeutic ideas for the pathogenesis,prevention and treatment of essential hypertension.Methods:Elderly patients aged 60 and above who underwent physical examination in shiwen town central hospital of fushun city,liaoning province and xingsheng community health service center of panjin city from January to October 2017 were selected as the study subjects,among which 431 patients with hypertension were selected as the case group and 345 healthy patients during the same period were selected as the control group.Compare the general epidemiological indicators of the two groups of subjects,real-time quantitative PCR was used to detect gene SNPs,and analyze the experimental results with a variety of genetic models and statistical methods.Results:The studies showed that alleles and genotypes of ZC3HC1 rs11556924 and VEGFA rs9472135 were not statistically different between the case group and the control group(P > 0.05).The allele of FGD5 gene rs11128722 was statistically different between the two groups(P=0.045).By gender stratification analysis,it was found that the allele carrying rate of FGD5 rs11128722 in the male population was statistically significant between the two groups(P=0.035).The genotypes of ZC3HC1 rs11556924,FGD5 rs11128722 and VEGFA rs9472135 were not significantly different in the additive,dominant and recessive genetic models(P > 0.05).Stratified by gender,there was no statistically significant difference between the genotype of ZC3HC1 gene rs11556924 and VEGFA gene rs9472135 between the case group and the control group(P > 0.05).Under the dominant genetic model,the genotype distribution of male FGD5 gene rs11128722 was statistically different between the two groups(P=0.049).FGD5 rs11128722 polymorphism had a statistically significant difference in body mass index among different genotypes(P=0.038).The body mass index of genotype GA in the additive genetic model was higher than that of GG genotype.Moreover,the body mass index of genotype GA+AA was higher than that of GG genotype in the dominant genetic model(P=0.011).Conclusion:In Liaoning province: the polymorphism of ZC3HC1 rs11556924 and VEGFA rs9472135 may not be significantly associated with the risk of primary hypertension;the polymorphism of FGD5 gene rs11128722 may play an important role in reducing the risk of essential hypertension,especially in elderly men.However,a larger sample size is still needed to confirm the results of this study.
