Objective:To investigate the effect of dexamethasone on sodium/iodide symporter?NIS?expression and iodine uptake in papillary thyroid carcinoma cells and thyroid follicular epithelial cells.Methods:Human papillary thyroid carcinoma cells?K1?and thyroid follicular epithelial cells?Nthy-ori3-1?were divided into two groups:control group without dexamethasone and experimental group with different concentrations of dexamethasone(10-9M,10-8M,10-7M and 10-6M),Western blot was used to detect the expression of NIS protein on the two kinds of cell membrane after 48 hours,and the gray ratio of NIS/?-actin band was used as the expression intensity of NIS protein.The two kinds of cells were cultured by the same grouping method for 48 hours,washed twice and then cultured in HBSS solution containing 185 kBq Na131I for 30 minutes.The radioactivity counts of the two kinds of cells were measured by?-counter for 60 seconds.One-way analysis of variance?ANOVA?was used to compare the differences groups and multiple comparisons were performed by LSD.Results:Dexamethasone at different concentrations down-regulated the expression of NIS protein in papillary thyroid carcinoma cells and thyroid follicular epithelial cells,which was statistically significant differences between groups?F=3.87,P<0.05?.However,there was no statistically significant differences in the expression of NIS protein in papillary thyroid carcinoma cells among different concentrations of dexamethasone of the experimental group,but there was significant difference in thyroid follicular epithelial cells between10-9M dexamethasone group and 10-6M dexamethasone group.Different concentrations of dexamethasone can affect the radioiodine uptake in papillary thyroid carcinoma cells and thyroid follicular epithelial cells obvious.The radioactive counts of both groups decreased with the increase of dexamethasone concentration,and there was statistically significant differences between groups?F values were 21.148,248.145,P<0.05?.The change of radioactive counts was a significant difference in papillary thyroid carcinoma cells between 10-9M dexamethasone group and 10-6M dexamethasone group.When the local dexamethasone concentration of papillary thyroid carcinoma cells reached 10-7M,there was no significant difference in radioactive count change in thyroid follicular epithelium after continued to increase in dexamethasone concentration.Conclusion:Dexamethasone can significantly reduce the expression of NIS protein and iodine uptake in human papillary thyroid carcinoma cells and thyroid follicular epithelial cells.
