Liver cancer is the sixth most common malignant tumor in the world,liver cancer is the third leading cause of cancer-related deaths in China.Among them,hepatocellular carcinoma(HCC)represents approximately 90% of all cases of primary liver cancer.Currently,transarterial chemoembolization(TACE)using ethiodised oil as an embolic agent is the most important palliative treatment for advanced hepatocellular carcinoma.However,the ethiodised oil emulsions used for the treatment of HCC prepared by conventional clinical pumping methods are often very unstable and unrepeatable.There is no doubt that unstable ethiodised oil emulsions cannot ensure effective clinical effect.In the research of this paper,a mixing device acting as a more accurate machine was designed to replace the conventional manual pumping process of preparing ethiodised oil emulsion,and on this basis,to study the improvement of ethiodised oil emulsion stability.In this study,the stability of the emulsion was studied by adding a certain amount of surfactant or biodegradable polymer particles,respectively.Among them,the effect of surfactant HCO-40 on the stability of ethiodised oil emulsion was systematically studied.The influence of factors such as preparation temperature,mixing times,emulsifier concentration,mixing ratio and other factors were investigated.The prepared ethiodised oil emulsions are stable for more than 24 hours,and has a certain drug sustained-release ability.Then,on this basis,the hydrophilic-lipophilic balance(hydrophilic-lipophilic balance,HLB)value was introduced to optimize the emulsifiers system,and it was found that the HLB value affected the stability of the emulsion.Similarly,thanks to Pickering technology,biodegradable particles are used to obtain a stable ethiodised oil Pickering emulsion.First,electrospinning technology and frozen milling were used to prepare irregularly shaped and large-sized polylactic acid fiber particles,which were used to study the effect of different hydrophilic and hydrophobic particles on the stability of the emulsions.Secondly,premix membrane emulsification was employed to prepare carboxyl-terminated polylactic acid nanoparticles of uniform and larger size.The influence of nanoparticles with different molecular weights on the stability of Pickering emulsion was investigated.The prepared emulsions are stable for more than 24 hours,accompanied with a certain drug sustained release ability.Finally,a translucent ex vivo model of a decellularized liver was constructed to visually monitor the distribution of Pickering emulsion in the liver vascular network after liver embolization in real time,and the feasibility of the model for evaluating the embolization effects of Pickering emulsion was preliminarily explored.
