Objectives Meta analysis was performed by reviewing relevant literature and cases to systematically evaluate the correlation between DRD3 gene Ser9 Gly polymorphism and essential tremor,so as to explore its pathogenesis and provide valuable reference for the study of ET genetics.Methods Wanfang database,Chinese National Knowledge Infrastructure,VIP database,Chinese biomedical literature database,Pub Med,Web of Science,Cochrance Library,Embase were searched by computer to collect case control studys on the correlation between DRD3 gene Ser9 Gly polymorphism and ET.Stata14.0 was used for meta analysis.Results A total of 8 case-control studies were included,with a total of 1456 cases and 1651 controls.The results of meta-analysis showed that there was no significant difference in the risk of ET between the G allele and the A allele in the allele model [OR=1.23,95%CI(0.95,1.59),P=0.116];there was no significant difference in the risk of ET between people with GG genotype and those with AA genotype in the homozygous model [OR=1.23,95%CI(0.87,1.75),P=0.232];there was no significant difference in the risk of ET between people with GA genotype and AA genotype in the heterozygous model [OR=1.14,95%CI(0.98,1.33),P=0.099];there was no significant difference in the risk of ET between people with GG genotype and those with GA+AA genotype in the recessive model [OR=1.16,95%CI(0.92,1.47),P=0.196];there was a significant difference in the risk of ET between people with GG+GA genotype and those with AA genotype in the dominant model [OR=1.16,95%CI(1.00,1.35),P=0.043].There was significant heterogeneity between allele model and homozygous model(P≤0.1,I2≥50%).The analysis of subgroups showed that the heterogeneity among the subgroups was significantly reduced based on race,suggesting that race may be an important factor in the origin of heterogeneity.In order to verify whether the relationship between Ser9 Gly polymorphism of DRD3 gene and the risk of ET is different among different human species,we also analyzed the heterozygote,recessive and dominant gene models based on race.The results showed that the risk of ET in the Caucasians of the dominant genotype was significantly different between the population with GG+GA genotype and the population with AA genotype(OR=1.20,95%CI(1.02,1.40),P=0.036),and the risk of ET in the population with GG+GA genotype was higher than that in the population with AA genotype.Conclusions Ser9 Gly polymorphism of DRD3 gene is associated with ET in Caucasian population,and the presence of GG+GA genotype may increase the risk of ET,but no correlation has been found in Asian population.Figure 12;Table 5;Reference 133...
