Objectives To study the effect of lncRNA CASC2 a on the proliferation,migration,invasion of human colon cancer cells,tumor angiogenesis,and its relationship with NF-?B signaling pathway.Methods Selected the cancer tissues and adjacent tissues of 43 patients with colon cancer from the affiliated hospital of North China University of Technology from October 2018 to January 2019,as well as human colon cancer cell lines HT-29,HCT-15,LOVO,COLO 205,SW480 and human The normal colonic epithelial cell line NCM-460,by RT-q PCR,was used to detect the relative expression levels of lncRNA CASC2 a in cancer tissues and adjacent tissues,as well as in five human colon cancer cells and human normal colon epithelial cells.LOVO colon cancer cells constructed lncRNA CASC2 a overexpression plasmid model,Western blot detection NF-?B signaling pathway related antibody protein p105 / p50,p65,tumor angiogenesis related antibody protein VEGF,MMP-9,MMP-2 expression,combined According to the previous experimental results,the expression of p105 / p50,p65 and VEGF,MMP-9,MMP-2 were detected after the stimulation of the NF-?B signaling pathway agonist propylene glycol methyl ether acetate(PMA)and compared with the previous ones.CCK-8,cell scratch test,and Transwell test were used to detect cell proliferation,migration,and invasion ability,respectively.Results The expression level of Lnc RNA CASC2 a in the cancer tissues of 43 patients with colon cancer is lower than that of the corresponding adjacent tissues(P<0.01),and the expression level in human colon cancer cells HT-29,SW480,HCT-15 is higher than that of human normal Colonic epithelial cells NCM-460,while the expression levels of COLO-205 and LOVO were lower than NCM-460(P<0.05),among which the expression level of LOVO was the lowest.After overexpression of lncRNA CASC2 a,the proliferation,migration,and invasion ability of cells in different groups of COLO-205 and LOVO weakened.The antibody protein p105 / p50 related to the NF-?B signaling pathway and the antibody proteins VEGF,MMP-9,MMP-2 related to tumor angiogenesis.After administration of the NF-?B signaling pathway agonist PMA,the ability to attenuate cell proliferation,migration,invasion,and the degree of inhibition of related antibody protein expression were all reduced.Conclusions LncRNA CASC2 a showed low expression in colon cancer tissues and COLO-205,LOVO colon cancer cells.Lnc RNA CASC2 a may inhibit the proliferation,migration,and invasion of human colon cancer cells COLO-205 and LOVO through the NF-?B signaling pathway;and inhibit the expression of tumor angiogenesis-related proteins VEGF,MMP-9,and MMP-2,thereby generating tumor metastasis Certainly affected.Figure 21;Table 26;Reference 138...
