The Effect Of Ke Min Qi Dan Nasal Spray On The Expression Of ECP And MiR-21 In The Nasal Mucosa Of Ar Rats With Qi Deficiency And Blood Stasis Syndrome

Objective:Through the treatment of allergic rhinitis model rats with Qi deficiency and blood stasis syndrome with Ke Min Qi Dan nasal spray,the effects of Ke Min Qi Dan nasal spray on the structure of nasal mucosa,hemorheology index,ECP level of nasal mucosa,expression level of miR-21 in serum,the mechanism of treatment and the dose effect relationship of the drug were discussed.From the aspect of basic experiment,we can lay the foundation and provide the basis for the clinical application of Ke Min Qi Dan nasal spray in the treatment of allergic rhinitis with Qi deficiency and blood stasis syndrome.Method:The model of AR rats with Qi deficiency and blood stasis syndrome was established by swimming exhaustion + cold dampness + high fat hunger combined with ovalbumin sensitization.It was divided into three groups: AR Qi deficiency and blood stasis syndrome control group,Kemin Qidan nasal spray high dose group,medium dose group,low dose group and Western Medicine group.In addition,a blank control group was set up.The Chinese medicine group was given the intervention of high sensitivity and low dose of Ke min Dan nasal spray.The western medicine group was given Mometasone Furoate Nasal Spray,the blank control group and the control group of AR Qi deficiency and blood stasis syndrome were not treated.The behavior and physical signs of rats were observed.After 14 days of administration,all rats were killed.The nasal mucosa was taken for HE staining and ECP protein content detection.The whole blood was taken for Hemorheology and miR-21 expression level detection.Statistical analysis of all experimental results.Result:Effects on the scores of behavior and physical signs: the scores of behavior and physical signs of the rats in each treatment group decreased after 7 days of administration,which were statistically different from those before administration and the control group of AR Qi deficiency and blood stasis syndrome;the scores of behavior and physical signs of the rats in the high dose group of traditional Chinese medicine and the western medicine group were the best,which were similar to each other.The influence on the change of hemorheology index: the model can increase the hemorheology index of rats,and cause blood stasis;after 14 days of treatment,the low cut,middle cut,plasma viscosity index and red blood cell aggregation index in the high dose group of traditional Chinese medicine were significantly different from those in the control group of AE Qi deficiency and blood stasis syndrome.The effect on the nasal mucosa cells of rats: the blank control group had no obvious pathological changes such as denatured cell necrosis;the control group of AR Qi deficiency and blood stasis syndrome had more serious infiltration,degeneration and death of mucosa epithelial cells,eosinophils infiltration,and the arrangement of connective tissue in the lamina propria was not tight;there were a few goblet cells in the high dose group of Chinese medicine,most of the cells had no obvious hyperplasia,degeneration or necrosis;In the middle dose group,the boundary of epithelial cells was not clear;in the low dose group,the boundary of epithelial cells was not clear,and there was inflammatory cell infiltration in the lamina propria;in the Western dose group,the boundary of epithelial cells was also unclear,with a little hyperplasia.Effect on the ECP level of nasal mucosa in rats: each treatment group has achieved significant therapeutic effect.Except for the comparison between the low dose group of traditional Chinese medicine and the AR Qi deficiency and blood stasis syndrome control group(P < 0.05),the ECP level of the high dose group of traditional Chinese medicine,the middle dose group of traditional Chinese medicine and the western medicine group has reached the significant difference level(P < 0.01),among which the ECP level of the middle dose group of traditional Chinese medicine is the same as that of the western medicine group,and the ECP content of the high dose group of traditional Chinese medicine is the lowest.Effect on miR-21 gene expression in rats: after 14 days of administration,the highest expression value of miR-21 in rats of each group was the high dose group of traditional Chinese medicine,and the lowest was the blank control group.The expression value ranged from high to low: high dose group of traditional Chinese Medicine > middle dose group of traditional Chinese Medicine > western medicine group > low dose group of traditional Chinese Medicine > ar Qi deficiency and blood stasis syndrome control group > blank control group.The expression level of miR-21 gene in the treatment group was statistically different from that in the AR Qi deficiency and blood stasis syndrome control group,except that in the low dose group of traditional Chinese medicine(P < 0.05),the rest were less than 0.01.Conclusion:1.Ke Min Qi Dan nasal spray can obviously relieve the symptoms of qi deficiency and blood stasis,such as depilation,loose stool,purple dark at the bottom of tongue,and the typical symptoms of allergic rhinitis,such as scratching nose,sneezing and runny nose;2.Ke Min Qi Dan nasal spray can reduce blood viscosity and relieve blood stasis in rats with allergic rhinitis of qi deficiency and blood stasis syndrome;3.Ke Min Qi Dan nasal spray can reduce the ECP protein content in the nasal mucosa of the allergic rhinitis rats with Qi deficiency and blood stasis syndrome,Light inflammation;4.Ke Min Qi Dan nasal spray can alleviate inflammatory cell infiltration and capillary dilation of nasal mucosa in rats with allergic rhinitis of qi deficiency and blood stasis syndrome,and alleviate the state of intranasal ischemia and hypoxia;5.Ke Min Qi Dan nasal spray can enhance the expression of miR-21 gene and promote the balance of Th1 / Th2 in the allergic rhinitis rats with Qi deficiency and blood stasis syndrome;6.Most of the above treatment levels in the low dose group were improved,but the improvement degree was limited;the middle dose group was effective,some of them reached the level of Western medicine group;the high dose group was effective,some of them exceeded the level of Western medicine group;7.The improvement degree of the above indexes can be improved with the increase of treatment time and concentration of traditional Chinese medicine in a certain range;8.The mechanism of Ke Min Qi Dan nasal spray in the treatment of allergic rhinitis of qi deficiency and blood stasis syndrome may be to promote the expression of miR-21 in rats,reduce the content of IL-4,inhibit the differentiation of Th0 to Th2,promote the balance of Th1 and Th2,and reduce the release of ECP in tissues,so as to alleviate the symptoms of rhinitis.
