| OBJECTIVE:1.To study the physicochemical properties of hot spring water in Mongolia Xiligalatuzhao 75 and Aohan hot spring in Inner Mongolia.2.Identify the genus and classification of the thermophilic flora of Mongolian Medical Hot Spring,purification and reproduction conditions,material metabolism,biological activity and dominant flora.3.Understand the method of isolating thermophilic bacteria,determine its medical effect,and provide a basis for future industry and medical treatment.Methods:1.Do systematic field surveys of Xirigala photos and Aohan hot springs,and use GPS to locate your location.2.Take the hot spring water of No.75 in Xirigala Photo,and use GB 8537-2008 and GB 8537-2016 to measure trace elements in the hot spring water.3.Separate and purify Xirigala Hot Spring and Aohan Hot Spring water on the thermophilic bacteria isolation medium,liquid expansion culture,extract DNA,16SrDNA for PCR amplification,and send its sequence to the company for testing.Blast analysis and comparison,phylogenetic analysis of the test results.Results 1.Mongolia’s Xirigala Hot Spring is located in Bayanhongor Province,650km away from the capital Ulaanbaatar.The geographical location is N:46.20’·E:101.14’,and the elevation is about 1492m.The temperature ranges from 15.7--82.6℃(self-test),PH8.2-8.9,which beongs to alkaline hot springs.There are more than 30 kinds of hot springs currently in use,and each hot spring has its own characteristics.China Aohan Hot Spring is located in Retang Village,Sijiazi Town,Aohan Banner,Chifeng City,Inner Mongolia Autonomous Region,China,150km away from Chifeng City.The geographical location is N:41.94’·E:119.96’,the altitude is about 695m,the temperature of Aohan hot spring is 68℃,PH7.47-7.77,alkaline hot spring.2.The chemical elements of Shargaljuut Hot Spring No.75 and Aohan Hot Spring are mostly the same.Shargaljuut Hot Spring is metasilicate hot spring and Aohan Hot Spring is radon hot spring.3.A total of 12 thermophilic bacteria isolated from Xarxil hot spring,Saahar hot spring and Ondegen hot spring in Shargaljuut,Mongolia,belong to three genera,among which bacillus and Anoxybcillus are dominant bacteria.Five thermophilic bacteria,three major genera,have been isolated from Aohan Hot Spring in Inner Mongolia,of which uncultured bacterium is the dominant genus.The phylogeny of the isolated thermophilic bacteria is dominated by the Bacillus group;it is divided into six genera,and the dominant bacteria are Anoxybacillus and Bacillus.Conclusion:1.Shargaljuut Hot Spring and Aohan Hot Spring stimulate the body through cold,heat,mechanical stimulation and trace elements,improve the body’s immunity,maintain the health of the body’s micronutrients,and regulate the body’s nine major systems.2.Compare the physicochemical properties of Shargaljuut’s NO75 hot spring and Aohan hot spring.The similarity is that both hot springs are alkaline hot springs.The hot spring water is clear and free of turbidity.The distribution of the four seasons is clear.Contains trace elements such as lithium,selenium,metasilicic acid,arsenic,copper,chromium,manganese,etc.Four microorganisms such as Escherichia coli,Streptococcus faecalis,difference is that the Shargaljuut Hot Springs are original hot springs.The hot springs and the surroundings of the hot springs are less artificially damaged than the Aohan Hot Springs.The hot springs flow directly from the original state.Shargaljuut photo NO:75 hot spring contains three elements such as nickel,pump acid and bromate.These three elements were not detected in Aohan hot spring.The unique elements of Aohan Hot Spring are radon and strontium.3.Isolation of thermophilic bacteria is an experiment in the scope of molecular experiments,which is a new challenge to the experimental research of Mongolian medical hot springs.4.The Shargaljuut Hot Spring and Aohan Hot Spring are unique hot springs of two countries,and further experimental research is needed to study its medical mechanism. |
