Objective:Explore the effect of long-chain non-coding RNA CECR7 on the proliferation and invasion ability of pancreatic cancer cells.Methods:The pancreatic cancer cell line PANC-1 cells were purchased from Shanghai Cell Bank,Chinese Academy of Sciences.Lentivirus infection technology was used to infect PANC-1 pancreatic cancer cells.Construct negative control lentiviral vector,CECR7 overexpression lentiviral vector,and infect PANC-1 pancreatic cancer cell line;the experiment is divided into three groups: blank control group(CON group): normal cultured cells without any interference measures;negative control group(NC group): Negative control lentiviral vector by infection;CECR7 overexpression group(CECR7 group): CECR7 overexpression lentiviral vector by infection.Collect the cells of each group after 48 hours of lentivirus infection,perform RNA extraction,reverse transcription,and fluorescent quantitative PCR to detect the expression level of CECR7 in the cells of each group,determine to establish a model of CECR7 overexpressing pancreatic cancer cells,observe the value-added Invasive ability.The value-added ability of cells was detected by MTT experiment method;the cell invasion ability in vitro was observed by Transwell experiment.Result: Real-time quantitative PCR results showed that CECR7 expression was significantly increased in the CECR7 overexpression group(CECR7 group)compared with the blank control group(CON group)and negative control group(NC group)(P <0.05),while the CON group There was no significant difference between CECR7 expression in the NC group,indicating that CECR7 overexpression pancreatic cancer cell model was successfully constructed.The results of MTT experiments showed that compared with the CON and NC groups,the cells in the CECR7 group significantly decreased in value at 24 h,48h,72 h,and96h(P <0.05),indicating that the cell growth was significantly inhibited after the expression of CECR7 was increased.Transwell experiments showed that compared with the CON and NC groups,the cell invasion ability in the CECR7 group was also significantly reduced(P<0.05),indicating that up-regulating CECR7 expression can inhibit the invasion ability of PANC-1 cells.Conclusion: CECR7 can inhibit the proliferation and invasion of pancreatic cancer cells,and is expected to become a new target for the treatment of pancreatic cancer,providing ideas and methods for the treatment of pancreatic cancer.
