Objective The purpose of this study was to establish the normal reference range of CD64 index of neutrophils in all age groups by flow cytometry,so as to provide clinical diagnostic basis.Methods A total of 766 subjects were included.After the outlier was removed,a total of 732(including 409 males and323 females)were included in this study.According to the documents issued by the American institute for clinical and laboratory standardization(CLSI)and the Chinese health industry standard WS/T 402-2012,the screening criteria were formulated,and they were divided into four groups according to the age.Group1: children group,1 ~ 12 years old(255 cases,including 148 males and 107 females);Group 2: adolescent group,aged between 13 and 18;Group 3: adult group,aged 19-59 years;Group 4: aged group ≥60years.BD FACS Caliburl flow cytometry was used to detect the neutrophil CD64 index,and the reference range of neutrophil CD64 index in ordos area was established.The influencing factors of neutrophil CD64 index were analyzed,and the established reference range was verified.Results A total of 766 subjects were included.After the screening criteria and outliers were eliminated,a total of 732(409 males and 323females)were included in this study.According to age groups,the first group was children(1~12 years old)(255 cases,including 148 males and 107 females).Group 2: adolescent group,aged from 13 to 18 years(128 cases,including 77 males and 51 females);Group 3: adult group,aged 19-59 years(215 cases,including 114 males and 101 females);Group 4: aged group: ≥60 years(134 cases,70 males and 64females).After excluding outliers,the sample quantity of each group was in line with the minimum number of 120 cases required by Chinese health industry standard WS/T 402-2012.When establishing the neutrophil CD64 index,no reference lower limit was set,and only the reference upper limit,namely 95%of the measured value,was taken.Gender and age differences were compared.It was found that there was no gender difference in the neutrophil CD64 index,there were significant differences in age,mainly between children aged 1~12 years old and adolescents,adult group,and elderly group.There was no statistically significant difference between adolescents,adult group and elderly group.Therefore,the adolescent group,adult group and elderly group were combined.The reference interval of CD64 index in ordos city was established: 1 ~ 12 years old CD64 index < 0.14;≥13 years old CD64 index <2.16.According to the standards of Chinese health industry,the established reference interval was verifiedand passed.Conclusion(1)the neutrophil CD64 index is independent of gender,so gender grouping is not necessary to establish the reference interval of neutrophil CD64 index.(2)there was a significant difference between the neutrophil CD64 index and age,mainly between children aged 1-12 years and other 3 age groups,and there was no statistically significant difference between the other 3 groups.(3)establish the reference interval of CD64 index in ordos region: 1 ~ 12 years old CD64 index ≤0.14;CD64 index ≥13years old≤ 2.16;(4)the established reference range of CD64 index of neutrophil cells was verified,which has been verified and can be applied to clinical practice.
