Objective: To study the effects of different concentrations of Schisandrin on the proliferation and apoptosis of thoracic aortic smooth muscle cells(vascular smooth muscle cell,VSMC)in rats incubated with high glucose,and to explore the potential efficacy of Schisandrin on the treatment of diabetic atherosclerosis(AS).Methods: Rat thoracic aortic smooth muscle cell line A7r5 was selected as the study object.They were divided into 6 groups: 1)normal glucose group: glucose(5.5mmol/L);2)high glucose group: glucose(25mmol/L);3)mannitol group: glucose(5.5mmol/L)+ mannitol(19.5mmol/L);4)high glucose + low dose schisandrin group: glucose(25mmol/L)+schisandrin(5?mol/L);5)high glucose+medium dose schisandrin group: glucose(25mmol/L)+schisandrin(10?mol/L);6)high glucose + high dose schisandrin group: glucose(25mmol/L)+schisandrin(20?mol/L).After 48 hours of cell culture,the expression of PCNA and Bax protein was detected by Western blot,the expression level of Bax protein was detected by immunocytochemistry,and the proliferation of A7r5 was detected by CCK-8 method.Results:1.Western blot results: Compared with NG group,the expression of PCNA protein increased in HG group,HG+L-S group,HG+M-S group and HG+H-S group(P<0.05);compared with HG group,the expression of PCNA protein decreased in NG group,OC group,HG+M-S group,HG+H-S group(P<0.05);compared with HG+L-S group,the expression of PCNA protein decreased in NG group,OC group,HG+M-S group and HG+H-S group(P<0.05).There was significant difference among HG+L-S group,HG+M-S group and HG+H-S group(P<0.05),and the expression decreased with the increase of drug concentration.Compared with NG group,the expression of Bax protein decreased in HG group,increased in HG+M-S group and HG+H-S group(P<0.05);compared with HG group,the expression of Bax protein increased in NG group,OC group,HG+M-S group and HG+H-S group(P<0.05);compared with HG+L-S group,the expression of Bax protein increased in HG+M-S group and HG+L-S group(P<0.05).There was significant difference between HG+L-S group,HG+M-S group and HG+H-S group(P<0.05),and the expression increased with the increase of drug concentration.2.Immunocytochemical results: Compared with NG group,the expression of Bax protein decreased in HG group,increased in HG+M-S group and HG+H-S group(P<0.05);compared with HG group,the expression of Bax protein increased in NG group,OC group,HG+M-S group and HG+H-S group(P<0.05);compared with HG+L-S group,the expression of Bax protein increased in HG+M-S group and HG+L-S group(P<0.05).There was significant difference between HG+L-S group,HG+M-S group and HG+H-S group(P<0.05),and the expression increased with the increase of drug concentration.3.CCK-8 detection results: OD value: Compared with NG group,HG group,HG+L-S group,HG+M-S group,HG+H-S group OD value was larger(P<0.05);compared with HG group,NG group,OC group,HG+M-S group,HG+H-S group OD value was smaller(P<0.05);compared with HG+L-S group,NG group,OC group,HG+M-S group,HG+H-S group OD value was smaller(P<0.05).There was significant difference among HG+L-S group,HG+M-S group and HG+H-S(P<0.05),and the OD value decreased with the increase of drug concentration.Conclusion:1.High glucose can induce the proliferation of VSMC and inhibit the apoptosis of VSMC.2.Schisandrin can inhibit the proliferation of VSMC in a high-glucose environment and promote its apoptosis in a dose-dependent manner,which plays a role in the prevention and treatment of diabetic AS.
