| Objectives To explore whether quinone dihydropterin reductase(qdpr)can affect renal interstitial fibrosis induced by ureteral obstruction(UUO)by regulating apoptosis.Methods 12 3-week-old SPF male SD rats were randomly divided into the following four groups by unilateral ureteral ligation and retrograde injection of QDPR lentivirus: sham operation group,UUO group,control group(UUO+QDPR empty recombinant lentivirus),QDPR high expression group(UUO+QDPR high expression recombinant lentivirus,QDPR+).The rats in each group were maintained for 14 days.The expression of QDPR,Type Ⅳcollagen,Vimentin,Bax,BCL2,protein in kidney of each group was detected by immunohistochemistry and Western method,and the deposition of collagen fibers was detected by picric acid complex red staining(VG).Results 1 The animal model of ureteral obstructive renal interstitial fibrosis induced by UUO was successfully established: The collagen deposition area was detected by VG staining in UUO and Sham groups,and the collagen deposition level was significantly increased in UUO group(P<0.05),while the expression of type IV collagen and Vimentin protein was significantly increased(P<0.05)and the expression of anti-apoptotic protein BCL2 was decreased in UUO group(P<0.05).2 After immunohistochemistry and Western validation,the overexpression model of QDPR protein in rat kidney was successfully constructed.3 Overexpression of QDPR alleviated UUO-induced renal interstitial fibrosis in rats by inhibiting apoptosis of renal tubules:VG staining showed that collagen deposition in QDPR+ group was significantly lower than that in no-load control group(P < 0.05).Immunohistochemistry and Western results showed that the expression levels of type IV collagen and Vimentin in QDPR+ group were significantly decreased(P<0.05).Apoptotic protein Bax was down-regulated(P<0.05)and anti-apoptotic protein BCL2 was increased(P<0.05).Conclusions 1 In UUO model,the expression of the anti-apoptotic protein BCL2 decreased,suggesting that UUO can induce apoptosis of renal tubular cells,and the expression of QDPR in UUO decreased,suggesting that it may play an important role in renal interstitial fibrosis.2 UUO-induced renal interstitial fibrosis in rats can be alleviated by overexpression of QDPR.Meanwhile,overexpression of QDPR inhibits apoptosis of renal tubules in rats,suggesting that QDPR may improve UUO-induced renal interstitial fibrosis in rats by inhibiting apoptosis.Figure5;Table2;Reference 249... |
