Objective:In order to explore the mechanism of Huanglian Jiedu Decoction to improve Aβ1-42 induced microglia inflammation,Aβ1-42 was used to intervene in microglia to simulate the inflammatory microenvironment in Alzheimer’s disease patients.Observe the changes in the expression of inflammatory factors,α7nAchR,and microglia phenotype-related proteins,and find out their internal connections,providing new ideas for the rational and effective use of related traditional Chinese medicine treatments to intervene in the progress of Alzheimer’s disease.Methods:Microglia in the logarithmic growth phase were randomly divided into 7 groups: negative control group,model group,blank control group,positive control group,Huanglian Jiedu Decoction group,Huanglian Jiedu Decoction plus MLA group,MLA group,respectively,without serum The culture medium,blank cerebrospinal fluid,and the corresponding concentration of drug-containing cerebrospinal fluid were added to the MLA group.The MLA solution with a final concentration of 10 μM was added.Except for the negative control group,oligomer Aβ1-42 with a final concentration of 5 μM was added to construct AD neuroinflammation Model,incubate for 24 hours.MTT was used to detect the damage of Aβ1-42 on microglia,and the effect of drug-containing cerebrospinal fluid in each group on the survival rate of microglia.The expression of TNF-α and IL-10 in BV2 cell supernatant was detected by ELISA.Western blot method was used to detect the expression levels of α7nAchR,i NOS and Arg-1 protein in BV2 cells.Use SPSS23.0 statistical analysis data to get the result.Results:1.Compared with the blank control group,1μM 20μM oligomer Aβ1-42 can reduce the cell survival rate;2.The drug-containing cerebrospinal fluid in each group had no significant effect on the survival rate of microglia;3.Huanglian Jiedu Decoction-containing cerebrospinal fluid down-regulates the expression of inflammatory factor TNF-α caused by oligomer Aβ1-42 in BV2 cells;upregulates the expression of IL-10,and its effect is inhibited after using blocking agents4.Huanglian Jiedu Decoction containing cerebrospinal fluid can reduce the expression level of i NOS protein in BV2 cells caused by oligomer Aβ and increase the expression level of Arg-1 protein in BV2 cells.Regulate the role of microglia phenotype.Conclusion:1.The oligomer Aβ1-42 can promote the activation of BV2 cells to M1 type and release the pro-inflammatory factor TNF-α.2.Huanglian Jiedu Decoction-containing cerebrospinal fluid can inhibit the proinflammatory factor induced by oligomer Aβ1-42,promote the release of antiinflammatory factor IL-10,induce M2 type activation of BV2 cells,and reduce the proinflammatory M1 type activation.This effect may be It is related to Huanglian Jiedu Tang’s ability to activate α7nAchR protein.
