Objective:Through the observation of the effect of Shenqi compound on the endothelial cells of high glucose "metabolic memory ",the mechanism of Shenqi compound inhibiting the metabolic memory of high glucose endothelial cells was discussed from the changes of cell proliferation,HIF-1 and IL-6 expression,which provided laboratory evidence for Shenqi compound to reduce vascular endothelial injury and protect blood vessels from diabetic complications.Methods:HUVEC were continuously cultured in high sugar(30 mmol/L)for 3 days,then transferred to normal sugar(5.5 mmol/L)for 4 days(resulting in high glucose "metabolic memory" cell model)after grouping: low concentration drug-containing serum group GT-L×3 days;medium concentration drug-containing serum group GT-M×3 days;high concentration drug-containing serum group GT-H×3 days;positive control group(resveratrol)GT-RV×3 days.Establishment of additional clusters,normal control group CON,D-glucose(5.5 mmol/L)×3 days;high glucose group GT,D-glucose(30 mmol/L)×3 days.The proliferation activity of each group endothelial cell was detected by CCK-8 method,the level of IL-6 was detected by ELASA method,and the expression of HIF-1 was detected by Q-PCR method.Results:1.Compared with the CON group,the proliferative activity of endothelial cell decreased significantly in the GT group(P<0.05);compared with the GT group,the torsion effect of the GT-M?GT-H and GT-RV groups on the inhibition of proliferation in endothelial cell was significant(P<0.05),showed a dose-dependent form to improve proliferation inhibition high glucose in "memory" cell,increase activity(P<0.05),and promote vascular endothelial repair.2.Compared with the CON group,the expression of HIF-1 in endothelial cell was significantly increased in GT group(P<0.05);compared with the GT group,theexpression of HIF-1 in endothelial cell was significantly decreased in GT-L?GT-M?GT-H and GT-RV groups(P<0.05);with the intervention of drug-containing serum,the expression of HIF-1 was gradually down-regulated and significantly inhibited(P<0.05),effect was significant at high concentrations of drug-containing serum(P<0.05).3.Compared with the CON group,the level of IL-6 of endothelial cell in GT-L?GT-M?GT-H?GT-RV group increased(P<0.05),but were lower than that in GT group(P<0.05);compared with the GT group,the level of IL-6 of endothelial cell in GT-L?GT-M?GT-H?GT-RV group decreased significantly(P<0.05),but all of them were higher than that in CON group(P<0.05).The glucose stimulated IL-6expression,and gradually returned to normal level with the intervention amount of traditional Chinese medicine(P<0.05),and the effect of high concentration drug-containing serum was obvious(P<0.05).Conclusion:1.Cell proliferation?HIF-1 and IL-6 are involved in the high glucose "metabolic memory" effect of vascular endothelial cell and intervene in the vascular endothelial injury mechanism.2.Shenqi compound can reduce the damage to vascular endothelial,achieve the role of protecting blood vessels to prevent DM vascular complications by increasing the proliferative activity,decreasing the expression of HIF-1,reducing the level of IL-6 in high-sugar "memory" injury cell.3.Resveratrol has the similar effect to Shenqi compound to improve the proliferative activity,down-regulate HIF-1 expression and reduce the IL-6 level in cell of high glucose "memory" injury,and then reduce the damage to vascular endothelial,achieving the function of protecting blood vessels and prevention and treatment of DM vascular complications.The effect is equivalent to high dose compound.
